摘 要:目的:探讨6,7-二甲氧基香豆素(6,7-DOC)对K562细胞红系分化的影响及作用机制。方法:选择K562细胞为研究对象,用联苯苄胺染色,考察1、5、10μmol·L^-16,7-DOC分别于0、24、48、72 h对K562细胞红系定向分化的联苯苄胺阳性率的影响;采用Western Blot检测不同浓度6,7-DOC诱导后K562细胞中红系分化标志蛋白α-globin、β-globin、γ-globin的表达变化;采用CCK-8和流式细胞术检测经6,7-DOC诱导后K562细胞的增殖和细胞周期分布情况。结果:5、10μmol·L^-16,7-DOC组联苯胺染色阳性率显著高于对照组,且呈时间与剂量依赖性;10μmol·L^-16,7-DOC组α-globin、β-globin、γ-globin的蛋白表达量显著高于对照组;5μmol·L^-1、10μmol·L^-16,7-DOC组两组细胞增殖率均显著低于对照组,且呈剂量、时间依赖性,6,7-DOC使K562细胞大量阻滞于G0/G1期,S期比例减少。结论:6,7-DOC能够抑制K562细胞增殖,使红系分化标志蛋白α-globin、β-globin、γ-globin表达增加,促进K562细胞向红系定向分化。Objective:To investigate the effect and its mechanism of 6,7-dimethoxycoumarin(6,7-DOC)on erythroid differentiation of K562 cells.Methods:K562 cells were selected as the study object and stained with biphenylbenzylamine.The effects of 1μmol·L^-1,5μmol·L^-1 and 10μmol·L^-16,7-DOC at 0 h,24 h,48 h and 72 h on the positive rate of biphenylbenzylamine staining in directional differentiation of K562 cells were investigated.The expression of erythroid differentiation marker proteinsα-globin,β-globin andγ-globin of K562 cells induced by different concentrations of 6,7-DOC were detected by Western Blot;Proliferation and cell cycle distribution of K562 cells induced by 6,7-DOC were detected by CCK-8 and flow cytometry.Results:6,7-DOC at 5μmol·L^-1 or 10μmol·L^-1 showed significantly higher positive rate of benzidine staining than the control,with time and dose dependence.α-globin,β-globin andγ-globin protein expression were significantly higher with 10μmol·L^-16,7-DOC than with control;The proliferation rates of cells in the 5μmol·L^-1 and 10μmol·L^-16,7-DOC groups were significantly lower than that in the control group,with dose-and time-dependence.Conclusion:6,7-DOC can inhibit cell proliferation and increase the expression of erythroid differentiation marker proteinsα-globin,β-globin andγ-globin,which promoted the erythroid differentiation of K562 cells.
