摘 要:目的观察山楂消脂胶囊对代谢综合征大鼠糖脂代谢的影响并探讨作用机制。方法采用高脂高糖高盐饮食诱导代谢综合征大鼠模型,将造模成功的32只大鼠随机分为模型组、二甲双胍组和山楂消脂组,另取10只正常大鼠为对照组,并给予相应的药物灌胃4周。第20周检测各组大鼠的体质量、Lee′s指数、腹腔脂肪质量、体脂比、收缩压(SBP)、舒张压(DBP)、空腹血糖(FBG)、空腹胰岛素(Fin)、总胆固醇(TC)、三酰甘油(TAG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、游离脂肪酸(FFA)、过氧化物酶体增殖剂激活受体-γ(PPAR-γ)、蛋白激酶(PKB)、磷脂肌醇-3激酶(PI3K)、脂联素(ADPN)、肿瘤坏死因子-α(TNF-α)、瘦素(LEP),并计算胰岛素抵抗指数(IRI)。结果与对照组比较,模型组大鼠的体质量、Lee′s指数、腹腔脂肪质量、体脂比、SBP、DBP、FBG、Fin、TC、TG、LDL-C、IRI、FFA、TNF-α、LEP水平明显升高(P<0.05),PPAR-γ、PKB、PI3K、ADPN水平明显降低(P<0.05);与模型组比较,山楂消脂组大鼠体质量、Lee′s指数、腹腔脂肪质量、体脂比、SBP、DBP、FBG、Fin、TC、TG、LDL-C、IRI、FFA、TNF-α、LEP水平明显降低(P<0.05),PPAR-γ、PKB、PI3K、ADPN水平明显升高(P<0.05)。结论山楂消脂胶囊可能通过调节PPAR-γ信号通路,上调ADPN的表达和下调LEP的表达改善代谢综合征大鼠的糖脂代谢和胰岛素抵抗。Objective:To explore the effect and possible mechanism of Shanzha Xiaozhi Capsules on glucose and lipid metabolism in rats with metabolic syndrome(MS).Methods:A high-fat,high-sugar,high-salt diet-in⁃duced metabolic syndrome rat model was used,and 32 rats successfully modeled were randomly divided into the model group,the metformin group,and the Shanzha Xiaozhi group,and another 10 normal rats were taken as the control group.All rats were given intragastric administration for 4 weeks.At 20 weeks,the body weight,Lee′s in⁃dex,abdominal fat mass,body fat ratio,systolic blood pressure(SBP),diastolic blood pressure(DBP),fasting blood glucose(FBG),fasting insulin(Fin),and total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C),free fatty acid(FFA),peroxisome proliferator-activated re⁃ceptor-γ(PPAR-γ),protein kinase(PKB),phosphoinositide-3 kinase(PI3K),adiponectin(ADPN),tumor necro⁃sis factor-α(TNF-α),leptin(LEP)of each group were detected,and the insulin resistance Index(IRI)was calcu⁃lated.Results:Compared with the control group,the body mass,Lee′s index,abdominal fat mass,body fat ratio,SBP,DBP,FBG,Fin,TC,TG,LDL-C,IRI,FFA,TNF-α,LEP levels of the model group increased significantly(P<0.05),and the PPAR-γ,PKB,PI3K,ADPN levels reduced significantly(P<0.05);compared with the model group,the body weight,Lee′s index,abdominal fat mass,Body fat ratio,SBP,DBP,FBG,Fin,TC,TG,LDL-C,IRI,FFA,TNF-α,LEP levels of the Shanzha Xiaozhi group reduced significantly(P<0.05),and the PPAR-γ,PKB,PI3K,ADPN levels increased significantly(P<0.05).Conclusion:Shanzha Xiaozhi Capsuless may improve glucose and lipid metabolism and insulin resistance in rats with metabolic syndrome by regulating PPAR-γsignaling pathway,up-regulating the expression of ADPN and down-regulating the expression of LEP.
