摘 要:目的探讨人参茎叶总皂苷对宫内生长受限(IUGR)大鼠的保护作用。方法将SD大鼠随机分为空白组、模型组和人参茎叶总皂苷组。建立妊娠期低蛋白饮食IUGR大鼠模型。人参茎叶总皂苷组孕中期灌胃给药(50 mg/kg)。在出生后第1天,测定胎鼠的新生仔鼠数量、新生仔鼠体重、脏器指数和血清生化参数、肝脏形态。流式细胞仪检测肝脏自噬标志物LC3和Beclin1。结果人参茎叶总皂苷能明显改善IUGR胎鼠的新生仔鼠数量、新生仔鼠体重、肝形态;可恢复血清SST、GHRH、GH、IGF-1、T3、T4水平及抗氧化指标(SOD、GSH、MDA)。人参茎叶总皂苷组中LC3和Beclin1明显降低。结论人参茎叶总皂苷能改善低蛋白引起的大鼠宫内生长受限,可能与增加甲状腺激素、GHRH/SST/GH、激活IGF-1//INS信号通路、抗氧化、减少自噬有关。Objective To investigate the protective effect of ginsenosides on intrauterine growth retardation(IUGR)in rats.Methods Sprague-Dawley rats were randomly divided into 3 groups:the control group,the model group and the ginsenosides group.IUGR rat model was built by feeding a low protein diet during pregnancy.Ginsenosides were given by gavage administration(50 mg/kg).The number,body weight,organ index and serum biochemical parameters of newborn mice were measured on the first day after birth.The autophagy marker LC3 and Beclin1 were determined by flow cytometry.Results Ginsenosides significantly improved the number,weight and liver morphology of newborn rats.The levels of serum SST,GHRH,GH,IGF-1,T3,T4,SOD,GSH and MDA were recovered.LC3 and Beclin1 were significantly reduced in the total saponins group.Conclusion Ginsenosides can improve intrauterine growth restriction induced by low protein in rats,which may be related to increased thyroid hormones,GHRH/SST/GH,activation of the IGF-1/INS signaling pathway,antioxidation and reduction of autophagy.
