摘 要:目的:探讨青藤碱对胃癌MGC803细胞迁移和侵袭的影响及机制。方法:将胃癌MGC803细胞随机分成4组:对照组、青藤碱300,600,1 200 mg·L-1组,采用划痕愈合实验检测细胞迁移率,Transwell实验检测迁移细胞数和侵袭细胞数,qRTPCR检测环氧合酶2(COX-2)、血管内皮生长因子(VEGF)、基质金属蛋白酶9(MMP9)、基质金属蛋白酶2(MMP2) mRNA表达,Western blot检测COX-2、VEGF、MMP9、MMP2蛋白表达。结果:青藤碱300,600,1 200mg·L-1组胃癌MGC803细胞迁移率、迁移细胞数、侵袭细胞数、COX-2、VEGF、MMP9、MMP2 mRNA和蛋白表达显著低于对照组(P<0.05或P<0.01)。结论:青藤碱抑制胃癌MGC803细胞迁移和侵袭,其机制与下调COX-2、VEGF、MMP9、MMP2表达有关。Objective: To investigate the inhibitory effect and underlying mechanism of sinomenine on the migration and invasion of gastric cancer MGC803 cells. Methods: The gastric cancer MGC803 cells were randomly divided into the following four groups: control group,and sinomenine 300,600 and 1 200 mg·L-1 groups. The mobility of MGC803 cells was tested by scratch healing test and the number of migrating and invasive MGC803 cells were tested by transwell test. The expressions of COX-2,VEGF,MMP9 and MMP2 mRNA of MGC803 cells were tested by qRT-PCR and the expressions of COX-2,VEGF,MMP9 and MMP2 of MGC803 cells were tested by Western blot. Results: Compared to those in the control group,the cell migration rate,number of migrating cells,number of invasive cells,mRNA and protein expressions of COX-2,VEGF,MMP9 and MMP2 in sinomenine 300,600 and 1200 mg·L-1 groups were significantly reduced( P<0.05 or P<0.01). Conclusion: Sinomenine inhibits the migration and invasion of gastric cancer MGC803 cells,and its mechanism may be related to the down-regulation of COX-2,VEGF,MMP9 and MMP2 expressions.
