摘  要:目的:考察钙联合淫羊藿及女贞子对低骨密度大鼠的改善作用。方法:3~4周龄断乳雌性大鼠80只随机分为8组:正常对照组、低钙对照组、碳酸钙Ⅰ组和Ⅱ组、钙联合淫羊藿Ⅰ组、钙联合女贞子Ⅰ组、钙联合淫羊藿及女贞子Ⅰ组和Ⅱ组,采用低钙饲料连续喂养造模,每周测量体质量及身长,喂养3周后进行钙代谢实验,测量摄入钙、粪钙含量,计算钙吸收率,取材前测量腰椎骨密度,取左侧股骨测量股骨折断力,计算股骨干重及骨钙含量,HE染色观察骨小梁结构;实时荧光定量逆转录聚合酶链反应(RT-PCR)法、蛋白质免疫印迹(Western Blot)法测定股骨中低密度脂蛋白受体相关蛋白5(LRP5)mRNA及蛋白表达。结果:与正常对照组相比,低钙对照组大鼠各指标差异均有统计学意义(P<0.05),表明模型成功。与低钙对照组相比,各干预组大鼠身长、摄入钙含量、粪钙含量、钙吸收率、腰椎骨密度、股骨最大折断力、左侧股骨干重及骨钙含量等差异均有统计学意义(P<0.05);与同剂量钙组相比,钙联合淫羊藿及女贞子Ⅰ组身长、钙吸收率的增加更显著(P<0.05)。与正常对照组相比,低钙对照组LRP5 mRNA表达及蛋白表达均显著下调(P<0.05);与低钙对照组相比,各干预组均可显著上调LRP5 mRNA表达及蛋白表达(P<0.05);与同剂量钙组相比,钙联合淫羊藿及女贞子Ⅰ组LRP5蛋白表达上调更显著(P<0.05),且钙联合淫羊藿及女贞子相比钙联合淫羊藿或女贞子,LRP5蛋白表达上调更显著(P<0.05)。结论:钙联合淫羊藿及女贞子具有增强骨密度的功能,其作用机制可能与上调LRP5表达有关。Objective: To investigate the ameliorative effect of calcium combined with Epimedium and Fructus Ligustri ligustri in rats with low bone density.Methods: Totally 80 weaning female rats at the age of 3-4 weeks were randomly divided into 8 groups: normal control group,low calcium control group,calcium carbonate Ⅰ and Ⅱ group,calcium combined with epimedium Ⅰ group,calcium combined with fructus ligustri lucidi Ⅰ group,calcium combined with epimedium and fructus ligustri lucidi Ⅰ and Ⅱ groups,and the rats were with low calcium continuous feeding to make low bone density model. The body quality and body length of rats were measured every week. After 3-week feeding,calcium metabolism experiment was performed to detect calcium intake and fecal calcium content,and the calcium absorption rate was calculated. The lumbar density was measured before drawing materials,and then the left femur was drawn to measure femoral fracture force,and femoral weight and bone calcium content were calculated. HE staining was used to observe the bone trabecular structure,and the mRNA and protein expressions of low-density lipoprotein receptor associated protein 5( LRP5) in femur were determined by real-time fluorescence quantitative reverse transcription polymerase chain reaction( RT-PCR)and Western blot. Results: Compared with the normal control group,the differences in all indices in the low-calcium control group was statistically significant( P<0.05),indicating the successful modeling. Compared with the low calcium control group,there were statistically significant differences in body length,calcium intake,fecal calcium content,calcium absorption rate,lumbar density,maximum fracture force of femur,weight of left femoral shaft and bone calcium content in each intervention group( P<0.05). Compared with the same dose of calcium group,calcium combined epimedium I group and fructus ligustri lucidi I group increased the body length and calcium absorption rate( P<0.05).
