摘 要:从德州卓奥水质净化有限公司的活性污泥中分离筛选出具有良好絮凝效果的菌株,并通过优化培养条件,研究其絮凝特性。通过稀释涂布和平板划线法筛选得到7株具有较强絮凝活性的菌株,对絮凝效果最好的菌株Z11进行形态、生理生化特性以及16S rRNA序列分析,初步鉴定其种属;并采用单因素实验及高岭土悬浊液絮凝沉降试验,优化菌株的培养条件,并初步研究菌株Z11的絮凝特性。初步鉴定菌株Z11为死谷芽孢杆菌(Bacillus vallismortis),在菌株培养液添加量3 mL、pH 5.0、温度30℃、转速220 r/min条件下,对4 g/mL高岭土悬浊液和24 g/L 1%氯化钙溶液混合液的絮凝率最高为96.66%。死谷芽孢杆菌Z11表现出良好的絮凝效果,具有应用污水处理的巨大潜力,有望应用于高效率的污水处理。Strains with good flocculant effects were isolated from the activated sludge of Zhuo Ao water purification Co.,Ltd in Dezhou and their flocculation characteristics were investigated by optimizing culture conditions.Seven strains with strong flocculation activity were screened by dilution and spread plate method and plate streaking method.A flocculat-producing strain Z11 was obtained with the best flocculating effect.The single factor experiment and Kaolin suspension flocculation sedimentation experiment were used to optimize the culture conditions,and the flocculation characteristics of strain Z11 were preliminarily studied.The strain Z11 was tentatively identified as Bacillus vallismortis based on its morphological and physiological-biochemical characteristics and 16S rRNA sequence analysis.The optimum culture conditions for Z11 were determined as follows:MBF dosage of 0.0048 g/L,pH 5,temperature 30℃,rate of shaking incubator 220 r/min,Kaolin concentration 4 g/mL,using 24 g/L of CaCl(mass concentration of 1%)as coagulant.Under the optimized conditions,the flocculating rate of Z11 reached as high as 96.66%.The results revealed that the Bacillus vallismortis Z11 showed good flocculation effect and had great potential for efficiency sewage treatment.
