摘  要:目的:观察茯苓多糖对复发性流产(RSA)小鼠妊娠结局的影响,并初步探讨其作用机制。方法:将雌性CBA/J小鼠与雄性DBA/2小鼠按照2:1比例交配,建立RSA模型,成功后随机分为模型组、茯苓多糖低、中、高剂量组和阳性对照组,另将雌性CBA/J小鼠和BALB/c小鼠按照2:1交配作为正常妊娠模型,设置为对照组。妊娠第1天,给予对照组和模型组蒸馏水灌胃处理,茯苓多糖低、中、高和阳性对照组依次给予使用蒸馏水溶解的50、100、200 mg茯苓多糖、156 mg/kg黄体酮胶囊灌胃处理,灌胃体积0.1 ml/10 g,每天1次,连续16天。观察并记录各组小鼠子宫组织胚胎着床数目和吸收数目以评估妊娠情况;计算各组小鼠脾脏和胸腺指数;流式细胞术检测各组小鼠子宫组织辅助性T淋巴细胞Th17、Treg、CD4+和CD8+T淋巴细胞比例;白细胞介素IL-17、IL-22、IL-10和TGF-β试剂盒说明书检测各组小鼠子宫组织上述各指标水平。结果:与对照组相比,模型组RSA小鼠胚胎丢失率、子宫组织Th17细胞比例、Th17/Treg比值、IL-17和IL-22表达、CD8+数量均显著升高,脾脏和胸腺指数、子宫组织Treg细胞比例、IL-10和TGF-β表达、CD4+数量、CD4+/CD8+比值均显著降低(均P<0.05);与模型组相比,茯苓多糖低、中、高剂量组RSA小鼠胚胎丢失率、子宫组织Th17细胞比例、Th17/Treg比值、IL-17和IL-22、CD8+数量依次降低,脾脏和胸腺指数、子宫组织Treg细胞比例、IL-10和TGF-β表达、CD4+数量、CD4+/CD8+比值依次升高(均P<0.05),阳性对照组上述指标变化趋势与茯苓多糖各组一致。结论:茯苓多糖能够改善RSA小鼠妊娠不良结局,可能与调节子宫组织Th17/Treg免疫细胞平衡有关。Objective:To observe the effect of pachyman on the pregnancy outcomes of mice with recurrent spontaneous abortion(RSA),and to explore its mechanism.Methods:Female CBA/J mice and male DBA/2 mice were mated according to the ratio of 2:1 to establish RSA model.They were randomly divided into model group,low,medium and high dose pachyman group,and positive control group.Female CBA/J mice and BALB/c mice were mated according to the ratio of 2:1 to establish normal pregnancy model in control group.On the first day of pregnancy,the female mice in control group and in model group were given distilled water by gavage,the female mice in the low,medium,high pachyman groups and positive control group were given 50,100,200 mg pachyman and 156 mg/kg progesterone capsules dissolved in distilled water by gavage,the volume of gavage was 0.1 ml/10 g,once a day for 16 consecutive days.The numbers of embryo implantation and absorption were observed and recorded to evaluate their pregnant situation.The spleen and thymus indexes of mice in these groups were calculated.The ratios of Th17,Treg,CD4+and CD8+T lymphocytes in uterine tissue of mice in these groups were detected by flow cytometry.And the levels of IL-17,IL-22,IL-10 and TGF-βwere detected by kits.Results:Compared with those of mice in the control group,the rate of embryo loss,the ratio of Th17 cells,the ratio of Th17/Treg,the expression of IL-17 and IL-22,and the number of CD8+of mice in the model group were significant higher,but the index of spleen and thymus,the ratio of Treg cells in uterus,the expression of IL-10 and TGF-β,the number of CD4+,and the ratio of CD4+/CD8+of mice in the model group were significant lower(all P<0.05).Compared with those of mice in the model group,the rate of embryo loss,the ratio of Th17 cells,the ratio of Th17/Treg,the expression of IL-17 and IL-22,and the number of CD8+of mice in low,medium and high dose pachyman groups decreased in turn(P<0.05),but the index of spleen and thymus,the ratio of Treg cells in uterus,the expression of IL-10 an
