梨试管苗种质资源保存的研究

摘　　要：本试验以黄花梨试管苗为试材,研究黄花梨离体培养保存,并筛选出最佳保存培养基。结果显示:最佳保存培养基为MS+6-BA(0.5 mg·L^-1)+IBA(0.1 mg·L^-1)+琼脂(6.0g·L^-1)+蔗糖(30 g·L^-1)+PP333(6 mg·L^-1)+甘露醇(5 g·L^-1)。以此为培养基,将梨10份种质资源进行离体试管苗保存,并对保存了6个月的10份种质资源的生长情况进行了观察。In this experiment,the test-tube plantlets of pear cv.‘Huanghua’were used as materials in vitro conservation to screen the best preservation medium.The results showed that the best preservation medium was MS+6-BA(0.5 mg·L^-1)+IBA(0.1 mg·L^-1)+agar(6.0 g·L^-1)+sucrose(30 g·L^-1)+PP333(6 mg·L^-1)+mannitol(5 g·L^-1).Ten accessions of pear were preserved in vitro on the above medium,and their growth preserved for six months was observed.
