当归补血汤与吉西他滨联用对肺癌A549细胞的抑制作用

摘　　要：目的研究当归补血汤联用吉西他滨对非小细胞肺癌A549细胞的生长抑制作用。方法比较水煎煮、水蒸气蒸馏、超声提取及索氏提取等不同制备方法对当归补血汤主要活性成分含量的影响,并考察对A549细胞的生长抑制作用,评价方法提取效率。MTT法评价当归补血汤中活性成分单体、当归补血汤提取物以及单体或提取物与吉西他滨联用后对A549细胞增殖活力的影响,考察当归补血汤与吉西他滨的相互作用。结果乙醇索氏提取当归后将黄芪与当归加水混合煎煮制备当归补血汤,可对水溶性和挥发油类活性成分进行高效提取。MTT结果显示,挥发油类成分蒿本内酯、阿魏酸松柏酯及当归补血汤均对A549细胞有明确生长抑制作用;挥发油类成分单体和当归补血汤分别与吉西他滨联用后,抑制肿瘤细胞生长作用均具有加和性,无明显协同作用;当归补血汤可抑制耐药蛋白转运体P-gp的活力。结论当归补血汤可抑制A549细胞增殖,但联用吉西他滨后无协同作用,为临床实践中运用当归补血汤辅助吉西他滨治疗非小细胞肺癌的机制研究提供了参考。Objective To study the growth inhibition effect of Danggui Buxue Decoction(DBD)combined with gemcitabine on non-small cell lung cancer(NSCLC)A549 cells.Methods Different extraction methods of preparation of DBD were compared with water decoction,steam distillation,ultrasonic extraction and soxhlet extraction.The main active ingredient contents and the growth inhibition effect on A549 cells were investigated.The effects of active component monomers,DBD extract and in combination with gemcitabine on the proliferation of A549 cells were evaluated by MTT method to investigate the interaction between DBD and gemcitabine.Results Among the different extraction methods,the extraction efficiency of soxhlet extraction was higher.Preparation of DBD was accomplished by soxhlet extraction combined with boiling water extraction to retain the main volatile and water-soluble active ingredients.Ligustilide,coniferate ferulate and DBD extract could inhibit the proliferation of A549 cells.The growth inhibitory effects of volatile oil component monomer and DBD combined with gemcitabine were additive,and no significant synergy was observed.Conclusion DBD can inhibit the proliferation of A549 cells,and the inhibition of tumor cells is additive after combination with gemcitabine.The obtained results provide important reference for using DBD as a new adjuvant therapy of gemcitabine for NSCLC.
