HPLC同时测定姜黄素胡椒碱前体脂质体中姜黄素和胡椒碱的含量

摘　　要：目的:通过高效液相色谱法(HPLC),建立同时测定姜黄素胡椒碱前体脂质体中指标成分(姜黄素、胡椒碱)含量的方法,进行相关系统适应性研究,并测定三批样品的姜黄素和胡椒碱的含量。方法:采用安捷伦C18色谱柱(型号Eclipse XDB,规格4.6×250mm,5μm),流动相为乙腈-1%磷酸(47:53),流速为0.8m L/min;柱温为25℃,检测波长分别为343nm(胡椒碱)和430nm(姜黄素)。结果:姜黄素的回归方程为y=89.956x+2.1408,r=0.9999,在0.56~17.86μg/m L之间线性关系良好,平均加样回收率为99.99%,RSD为0.64%。胡椒碱的回归方程为y=83.390x+0.7363,r=0.9999,在0.43~13.82μg/m L之间线性关系良好,平均加样回收率为100.68%,RSD为0.78%。结论:本法快速、操作简便,供试品色谱图主成分峰峰形对称,分离度高,方法的专属性好、重复性好、准确度高,可用于姜黄素胡椒碱前体脂质体中姜黄素和胡椒碱的含量测定。Objective:To establish a HPLC method for determining simultaneously the contents of Curcunin and Piperine in Compand curcunin-piperine Proliposomes.Methods:HPLC was used to determine simultaneously the contents of Curcunin and Piperine in the Compand curcunin-piperine Proliposomes.Agilent Eclipse XDB-C18 column(4.6 mm×250 mm,5μm)was used with a step gradient of Acetonitrile-1%phosphoric acid(47:53)at a flow rate of 0.8 mL/min.The detection wavelength of Curcunin was set at 343 nm,meanwhile the detection wavelength of Piperine was set at 430 nm.The column temperature was at 25℃.Results:There was good linear relationship in the range of 0.56~17.86μg/ml for Curcunin and 0.43~13.82μg/ml for Piperine.The correlation coefficinets of Curcunin and cinnamaldehyde were higher than 0.999.The average recovery of Curcunin was 99.99%(n=9),and Piperine was 100.78%(n=9).RSD of the above two components were 0.64%and 0.78%respectively.Conclusion:The method is simple,accurate and can be used as the method for quality assessment of Compand curcunin-piperine Proliposomes.
