摘 要:选取6周龄SPF级SD大鼠,随机分空白对照组(阴性对照组)、阳性对照组(不投用药物治疗)、筋骨七厘散低剂量治疗组(0.15 g/kg)、筋骨七厘散中剂量治疗组(0.30 g/kg)、筋骨七厘散高剂量治疗组(0.60 g/kg),阳性对照组和治疗组实验大鼠均做股骨骨折模型,治疗组用药为1次/d,连续用药10 d。试验期14 d,试验期间实验大鼠自由采食、饮水,试验期间观察实验大鼠临床体征。在试验第7天、第14天,分别检测实验大鼠血清骨形态发生蛋白(BMP)、β-转化生长因子(TGF-β)、胰岛素样生长因子(IGF-1)与肿瘤坏死因子(TNF-α)含量。试验结果表明,在实验大鼠骨愈合过程中,筋骨七厘散低剂量、中剂量、高剂量均能显著影响骨折实验大鼠血清BMP、TGF-β、IGF-1和TNF-α含量。投服0.30 g/kg、0.60 g/kg的筋骨七厘散影响效果最显著且2组间无明显差异,但2组效果均明显优于低剂量筋骨七厘散组(0.15 g/kg)。说明中药复方筋骨七厘散能调节骨折实验大鼠血清BMP、TGF-β、IGF-1与TNF-α含量变化,有效地促进实验大鼠骨愈合,且其疗效与投药量有一定关系。Six week old SPF SD rats were randomly divided into blank control group(negative control group),positive control group(no drug treatment),Jinguqili powder low-dose treatment group(0.15 g·kg-1),Jinguqili powder medium dose treatment group(0.3 g·kg-1)The experimental rats in the positive control group and the treatment group were all made femoral fracture models.The treatment group was given medicine once a day for 10 days.The experimental period was 14 days.During the experiment,the rats were free to eat and drink water.The clinical signs of the experimental rats were observed during the experiment.The levels of bone morphogenetic protein(BMP),transforming growth factor-β(TGF-β),insulin-like growth factor(IGF-1)and tumor necrosis factor(TNF-α)in serum were detected on the 7th and 14th day of the experiment.The results showed that Jinguqili powder could significantly affect the contents of serum BMP,TGF-β,IGF-1 and TNF-αin experimental rats during bone healing.The effect of 0.3 g·kg-1 and 0.6 g·kg-1 Jinguqili powder was the most significant,and there was no significant difference between the two groups,but the effect of the two groups was significantly better than that of the low-dose(0.15 g·kg-1)Jinguqili powder group.Jinguqili powder can regulate the changes of serum BMP,TGF-β,IGF-1 and TNF-αcontents in experimental rats with fracture,and effectively promote bone healing of experimental rats,and its curative effect is related to the dosage of medicine.
