摘　　要：目的为了明确味连须在治疗剂量或治疗剂量以上范围对生理机能的潜在不良作用,确定味连须可能关系到动物的安全性的非期望药理作用以及评价味连须在毒理学和/或临床研究中所观察到的药物不良反应和/或病理生理作用及其作用机理,开展了味连须的安全药理(SP)研究。方法试验通过采用生物机能实验系统(BL-420E+)对不同时间段给药后试验家兔的动脉血压、左心室内压、呼吸和心电图进行测定;而后采用转棒坠落法拟观察味连须对小鼠协调运动功能的影响;为记录药物对试验小鼠阈下剂量的戊巴比妥钠催眠作用,试验以翻正反射消失法进行相关测定。结果味连须对小鼠中枢神经系统(CNS)没有明显影响。在1.0、2.0、4.0g/kg三个剂量时对家兔呼吸系统及心血管系统指标均无明显影响。结论味连须的使用是安全的。Objectives In order to investigate possible adverse effects of Coptis chinensis Franch radix within or over therapeutic dose range,determine unwanted pharmacologic actions that may cause safety issues in animals,and evaluate the observed adverse effects and/or pathophysiological role as well as mechanism of action of Coptis chinensis Franch radix in toxicology and/or clinical studies,we carried out safety pharmacology(SP)studies in Coptis chinensis Franch radix.Methods The arterial pressures,left ventricular pressure,respiratory curve and electrocardiogram in rabbits were recorded before and after ig administration using BL-420E+multifunctional signal processor.The effects on coordinated movements of mice were tested by recording turn-club falling times.The drug’s influence on hypnosis induced by subthreshold dose of pentobarbital sodium was tested by observing the disappearance of righting reflex.Results Ig administration of Coptis chinensis Franch radix did not affect the central nervous system of mouse.Similarly,ig administration of Coptis chinensis Franch radix at 1.0,2.0,or 4.0g/kg dosage had no significant effect on the respiratory and cardiovascular systems of rabbit.Conclusion Coptis chinensis Franch radix is safe to use.
