摘 要:试验旨在探究猪卵母细胞和孤雌激活胚胎在运输箱内培养对卵母细胞体外成熟效率及后续胚胎发育潜能的影响。首先测定了运输箱的保温效果。结果显示,无论是否外界电源,置于该运输箱恒温浴内的1.5 mL、0.5 mL离心管和0.25 mL的麦管均可保持温度恒定。其次,将在常规培养箱体外培养的卵丘卵母细胞(COCs)设为对照组(A),分别将COCs置于1.5 mL的离心管(B)、0.5 mL的离心管(C)和0.25 mL的麦管(D)在运输箱中进行培养。体外培养44 h后,观察不同成熟条件下COCs卵丘细胞扩散情况并统计卵母细胞成熟率。结果表明,尽管在0.5 mL离心管中成熟的COCs卵丘细胞扩散情况相对较差,但无论在何种条件下(A,B,C和D),卵母细胞死亡率、成熟率均无显著差异。另外,分别从4组中挑选含有第一极体(PB1)的卵母细胞进行孤雌激活,并再次于对应的4种条件下(A-A′,B-B′,C-C′,D-D′)进行体外培养。胚胎体外发育的结果显示,4组胚胎在4-细胞时期分裂率无显著差异。温度恒定的运输箱条件能满足COCs完成体外成熟,且能支持胚胎完成至少初始的两次卵裂。In order to investigate the effects of porcine cumulus-oocyte complexes(COCs)and parthenogenetic activation(PA)embryos cultured in the delivery case on the efficiency of maturation and subsequent embryonic development potential,the temperature change in the delivery case was measured and then the cumulus-oocyte complexes(COCs)were cultured in the delivery case using 1.5 mL Eppendorf tubes(B),0.5 mL Eppendorf tubes(C)and 0.25 mL straws(D),The COCs cultured in the incubator(A)were used as the control group.The results showed that the delivery case maintained a constant temperature.In addition,although the cumulus expansion in Group C was not as good as in the other groups,no difference of the survive rate and the mature rate were observed among these four groups.Then,the early developmental ability of PA embryos was further investigated under the same condition as that of the oocyte maturation culture(A-A′,B-B′,C-C′,D-D′).The results showed that there was no significant difference in the 4-cell cleavage rate.In conclusion,oocytes could be matured and the PA embryos could developed into the 4-cell stage in the delivery case at appropriate temperature.
