摘  要:为调查新疆部分地区规模化牛养殖场牛病毒(BVDV)性腹泻病毒的感染情况,本试验采用RT-PCR方法对640份牛全血进行牛病毒性腹泻病毒检测,并对检测基因序列进行分析及同源性比较。结果表明:各地区牛养殖场BVDV阳性检出率为22.5%~62.5%,平均总阳性率为40.0%(256/640),不同年龄段牛群感染BVDV的平均阳性率为22.5%(27/120)。将目的片段克隆至T载体,经测序获得5株BVDV部分基因序列,与预期的大小一致为288 bp。根据同源性分析表明,本研究的5株克隆毒株序列之间的同源性为98.3%~99.7%。遗传演化分析可知,本研究获得的毒株在同一簇,与中国北京株BVDV1a亚型归为同一簇,亲缘关系比较接近,但与BVDV2型、BVDV3型的遗传关系较远。研究结果表明,新疆地区BVDV普遍存在,经测序核苷酸序列之间的同源性较高,遗传关系较接近,该结果为新疆地区牛病毒性腹泻病毒病的防控工作奠定基础。In order to fully understand the prevalence of bovine viral diarrhea(BVD)on large-scale cattle farms in Xinjiang,a total of 640 samples of bovine whole blood were collected and detected by RT-PCR.Then,the sequence and homology of the detected genes were analyzed.The results showed that the positive detection rates of BVDV ranged from 22.5%to 62.5%on the individual cattle farms in different regions,and their average total positive rate was 40.0%(256/640).The average positive rate of BVDV infection in cattle at different ages was 22.5%(27/120).The target fragment was cloned into the T vector,and partial gene sequences of the five BVDV strains were obtained,which was 288 bp as expected.According to the homology analysis,the homology of the five strains was 98.3%-99.7%.The genetic evolution analysis showed that the virus strains obtained in this study were in the same cluster,and they belonged to the same cluster with that of the BVDV1 a subtype of the Beijing strain in China,with a close genetic relationship,but they were of far genetic relationship with the BVDV2 and BVDV3 subtypes.The present results indicated that bovine viral diarrhea virus was prevalent in the Xinjiang region,and there was a high homology and close genetic relationship among the nucleotide sequences,which provided fundamental information of prevention and control of bovine viral diarrhea virus disease in Xinjiang Yugur Autonomous Region.
