二仙汤对去势大鼠骨微结构及ALP、OPG、IL-6的影响

摘　　要：目的研究二仙汤防治骨质疏松症的机制,探讨"温肾补精,以生髓壮骨"中医理论在绝经后骨质疏松症中的应用。方法采用卵巢切除法复制绝经后骨质疏松症大鼠模型,随机分为模型组、二仙汤组、雌二醇组,同时设置假手术组作为对照,各组分别给予相应的药物灌胃。给药3个月后处死取材,计算子宫系数;HE染色观察子宫的病理变化;Micro-CT检测大鼠骨密度及骨微结构的变化;全自动生化分析仪检测各组大鼠血清中血糖、血脂及钙磷的变化;ELISA检测各组大鼠血清中碱性磷酸酶(alkaline phosphatase,ALP)、白介素6(Interleukin,IL-6)及骨保护素(osteoclastogenesis inhibitory factor,OPG)表达的变化。结果与假手术组比较,模型组大鼠骨密度显著下降(P<0.05),骨微结构破坏明显;与模型组比较,二仙汤组大鼠骨密度得到了有效提高、骨微结构获得改善,血糖、血脂水平降低,血清中钙磷的含量明显提高,血清中ALP、IL-6含量降低,OPG含量升高(P<0.05,P<0.01)。结论二仙汤能够提高去卵巢大鼠的骨量,抑制破骨细胞活性,促进骨形成防治绝经后骨质疏松症,其作用机理可能与降低ALP、IL-6,提高OPG含量有关。Objective To study the mechanism of Erxian Decoction in preventing and treating osteoporosis,and to explore the application of TCM theory of"warming the kidney to supplement essence and giving birth to marrow and bone"in postmenopausal osteoporosis.Methods Postmenopausal osteoporosis rats were induced by ovariectomy.The rats were randomly divided into model group,Er Xian Tang group,estradiol group,and sham-operated group as control group.After3 months of administration,samples were collected and the uterus coefficient was calculated;pathological changes of uterus were observed by HE staining;changes of bone mineral density and bone microstructure were detected by Micro-CT;changes of blood glucose,blood lipid and calcium and phosphorus in serum were detected by automatic biochemical analyzer;changes of ALP,IL-6 and OPG expression in serum were detected by ELISA.Results Compared with the sham operation group,the bone density of rats in the model group was significantly decreased(P<0.05),and the bone microstructure was obviously destroyed;compared with the model group,the bone density of the rats in the Erxiantang group was effectively increased and the bone microstructure was obtained.Improved,blood sugar and blood lipid levels were reduced,serum calcium and phosphorus content was significantly increased,serum ALP,IL-6 content was reduced,and OPG content was increased(P<0.05,P<0.01).Conclusion Erxian Decoction can increase bone mass and prevent postmenopausal osteoporosis in ovariectomized rats.Its mechanism may be related to decreasing ALP,IL-6 and increasing OPG content.
