摘  要:为检测目前暂未有报道的小鼠下丘脑内3β羟基类固醇脱氢酶1(3β-HSD1)和细胞色素P45017A1(CYP17A1)的分布及表达,了解其在下丘脑调节激素分泌中的潜在作用。分别使用免疫荧光及Western Blot技术检测小鼠下丘脑内3β-HSD1和CYP17A1的神经元分布及蛋白表达情况。免疫荧光结果显示,3β-HSD1阳性神经元分布于下丘脑背外侧核、腹内侧核以及弓状核,CYP17A1阳性神经元分布于下丘脑腹内侧核及室旁核;Western Blot结果显示,下丘脑内均有3β-HSD1和CYP17A1蛋白表达。Western Blot试验证实,免疫荧光检测到下丘脑内3β-HSD1和CYP17A1分布的真实性。本试验首次检测出下丘脑内类固醇激素合成关键酶3β-HSD1和CYP17A1的免疫阳性神经信号的分布,为进一步研究下丘脑调节激素分泌提供形态学依据。In order to detect the distribution and expression of 3β-hydroxysteroid dehydrogenase 1(3β-HSD1)and cytochrome P45017A1(CYP17A1)in the hypothalamus of mice,and to understand their potential role in the regulation of hormone secretion in the hypothalamus.Immunofluorescence and Western Blot were used to detect the distribution and protein expression.Immunofluorescence showed that 3β-HSD1 positive neurons were distributed in the dorsolateral nucleus,ventromedial nucleus and arcuate nucleus of the hypothalamus,and CYP17A1 positive neurons were distributed in the ventromedial nucleus and paraventricular nucleus of the hypothalamus,Western Blot showed that 3β-HSD1 and CYP17A1 protein were expressed in the hypothalamus.Western Blot test confirmed the authenticity of the distribution of 3β-HSD1 and CYP17A1 in the hypothalamus detected by immunofluorescence.The distribution of immunoreactive nerve signals of 3β-HSD1 and CYP17A1,the key enzymes of steroid hormone synthesis in the hypothalamus is detected for the first time in this study,which provides morphological basis for further study of hypothalamic regulatory hormone secretion.
