摘 要:目的:探讨肉苁蓉总苷(GCs)对β-淀粉样蛋白致Alzheimer病(AD)大鼠海马CA1区神经元病理形态学的影响。方法:将60只SD雄性大鼠随机分为6组,分别为假手术组、β淀粉样蛋白组(模型组)、维生素E阳性药组(Vit E组)、GCs 40 mg/kg组(小剂量组)、GCs 80 mg/kg组(中剂量组)、GCs 160 mg/kg组(高剂量组)。灌胃给药,1次/d,连续14 d。给药结束后,处死大鼠,制备其脑组织切片备用。经苏木精-伊红染色(HE)和Bielschowski s银染色法观察海马CA1区神经元的病理结构改变,且通过电子显微镜观察神经元超微结构变化。结果:(1)GCs给药组的大鼠海马CA1区细胞排列整齐,形态正常,未见凋亡坏死细胞,结构与假手术组类似。(2)GCs各给药组的大鼠神经元细胞核圆,核膜完整,核仁明显,线粒体结构完整,神经原纤维排列较整齐,结构几乎接近于假手术组和维生素E阳性药对照组。(3)GCs给药组的大鼠海马CA1区神经原纤维排列较规则,神经元结构较完整,无明显肿胀、增粗,损伤明显减轻,组织结构基本接近于假手术和维生素E阳性药对照组。结论:GCs能减轻AD模型大鼠受损神经细胞的病理性改变及超微结构的损伤。Objective:To explore the effects of Glycosides of Cistanche(GCs)on the pathomorphology of neurons in the hippocampal CA1 area of Alzheimer s disease(AD)rats induced byβ-amyloid.Methods:60 male SD rats were randomly divided into 6 groups:sham operation group,amyloid beta group(model group),vitamin E positive drug group(Vit E group),GCS 40 mg/kg group(low dose group),GCS 80 mg/kg group(medium dose group)and GCS 160 mg/kg group(high dose group).The drug was given by gavage once a day for 14 days.After the administration,the rats were sacrificed and their brain tissues were prepared.The hematoxylin-eosin(HE)staining and Bielschowski s silver staining were used to observe the pathological structures of neurons in the CA1 region of the hippocampus.Also,the ultrastructural changes of neurons were observed by transmission electron microscope.Results:(1)The hippocampal CA1 region of the GCs group were normal,and no apoptotic necrotic cells were observed,which was similar to that of the sham operation group.(2)The neuronal nuclei of rats in each GCs administration group were round and their nuclear membranes were complete.Besides,their nucleoli were obvious and mitochondrial structures were complete.Moreover,their neurofibrils were arranged neatly and the structures were almost similar with that in the sham operation group and vitamin E positive group.(3)The neurofibrils in the hippocampal CA1 region of the rats in the GCs administration group were arranged regularly and their neuron structure was complete,indicating that there was no obvious swelling,thickening and the damage was significantly reduced.Additionally,their structure of tissue was basically close to the sham operation and vitamin E positive group.Conclusion:GCs reduce the pathological changes and ultrastructural damage of injured nerve cells in AD rats.
