摘 要:目的建立栀子、炒栀子、焦栀子的UPLC指纹图谱,结合多元统计分析评价不同炮制品的差异。方法应用Waters超高效液相色谱仪(UPLC)进行测定,色谱柱为YMC Triart C18(100 mm×2.1 mm,1.9μm)色谱柱,流动相为乙腈-水梯度洗脱,流速为0.4 mL/min,柱温为30℃,检测波长:0~6 min为238 nm,6~11 min为440 nm,11~18 min为238 nm。使用《中药色谱指纹图谱相似度评价系统》(2012年版)建立栀子、炒栀子与焦栀子的共有模式,并进行相似度评价。采用SIMCA-P 12.0软件进行正交偏最小二乘法判别分析(Orthogonal partial least square discriminant analysis,OPLS-DA),寻找对不同炮制品分组贡献最大的共有组分。结果3种炮制品共有7个共有峰,指认了其中的京尼平龙胆双糖苷、栀子苷、西红花苷-Ι、西红花苷-Ⅱ4个化合物,各炮制品具有一定的相似度。OPLS-DA结果发现栀子与焦栀子、栀子与炒栀子、焦栀子与炒栀子均存在差异,对差异贡献最大的组分分别是京尼平龙胆双糖苷与西红花苷-Ι、西红花苷-Ι与西红花苷-Ⅱ、西红花苷-Ι与西红花苷-Ⅱ。结论本方法适用于栀子不同炮制品的指纹图谱分析,能有效区分栀子的不同炮制品,为栀子不同炮制品的质量标准提升提供一定的参考价值。Objective The fingerprints of gardeniae fructus,stir-baked gardeniae fructus and gardeniae fructuspreparatus were established,and the difference between processed products was evaluated by multivariate statistical analysis.Methods Ultra Performance Liquid Chromatography(UPLC)was performed on a YMC Triart C18(100 mm×2.1 mm,1.9μm)column with acetonitrile-water as mobile phase in gradient mode at the flow rate of 0.4 mL/min.The column temperature was 30℃.Detection wavelength:238 nm for 0-6 min,440 nm for 6-11 min,and 238 nm for 11-18 min.The similarity evaluation System of Traditional Chinese Medicine Chromatographic Fingerprint(2012 edition)was used to establish the common pattern of gardeniae fructus,stir-baked gardeniae fructus and gardeniae fructuspreparatus,and the similarity evaluation was carried out.Orthogonal partial least square discriminant analysis(OPLS-DA)was performed to find the common components which contributed the most to different batches of processed products by SIMCA-P 12.0 software.Results There were 7 common peaks of raw gardenia,caramel gardenia and sauted gardenia.The gentiobioside,geniposide,crocinⅠ,crocinⅡwere identified respectively.They had high similarity among all products.OPLS-DA found that there were significant differences between gardeniae fructus and gardeniae fructus preparatus,gardeniae fructus and stir-baked gardeniae fructus,gardeniae fructuspreparatus and stir-baked gardeniae fructus,The group contribution rate of three components were gentiobioside and crocinⅠ,crocinⅠand crocinⅡ,crocinⅠand crocinⅡ.Conclusion The method is applicable to the fingerprint analysis of different processed gardenia,which can effectively distinguish different processed gardenia,and provide certain reference value for the quality standard improvement of different processed gardenia.
