摘 要:目的筛选可用于比格犬遗传检测的微卫星位点,并对小型比格犬群体进行了遗传结构分析。方法选取来自文献的微卫星位点120个,包括比格犬位点23个和其他犬类遗传检测的位点97个。用DNA池对这些位点进行PCR条件优化后,挑选扩增效果优良的位点对常用比格犬群体进行荧光标记STR扫描和群体遗传结构分析,依据期望杂合度(He)和香农指数挑选符合要求微卫星位点计算群体遗传参数。结果在候选的120个位点中共有73个位点扩增效果优良,STR扫描后选用49个位点进行遗传分析,结果显示该群体有效等位基因数、期望杂合度、平均杂合度、香农指数、多态性信息含量分别为4.9230、0.7574、0.7375、1.6625、0.7038,证明该群体遗传多态信息丰富,群体遗传多样性高。结论所选49个微卫星位点可用于比格犬遗传检测,这些位点还需用更多群体进行验证和优化。Objective In order to select microsatellite loci suitable for genetic detection of the outbred beagle dogs and to primarily adapt to analyze the genetic structure of one outbred population of small beagle dogs.Method We selected 120 microsatellite markers included with 23 loci from beagle dogs and 97 loci from other canine strains.The PCR conditions of these loci were optimized by DNA pool.The loci of better amplification effect were selected to analyze further with fluorescence labeled STR scanning and population genetic structure.Result A total of 73 loci were successfully amplified,within which 49 loci were used to calculate genetic information after STR scanning.It was shown that the small beagle dog population exhibited the effective number of allele 4.9230,expected heterozygosity 0.7574,average heterozygosity 0.7375,Shannon’s information index 1.6625 and PIC0.7038,respectively.It showed that the examined population exhibited high and abundant population genetic polymorphism information and high genetic diversity.Conclusion The set of 49 microsatellite loci we optimized could be adapted for genetically monitoring outbred beagle dogs.These loci are valuable to be verify and optimize using more populations.
