摘  要:目的对现有Hartley豚鼠种群进行剖腹产净化,通过遗传检测,确认净化后的种群符合封闭群遗传特征。方法生产群中不同繁殖单元广泛选择雌、雄个体,按1∶1比例合笼交配,于合笼后70 d实施剖腹产手术,仔鼠人工乳饲喂。用循环交配法繁殖一代,从F1代中选取30个个体采集耳部组织样本,提取DNA,应用18个豚鼠基因组微卫星位点进行遗传检测。结果剖腹产共获得85只仔鼠,成活56只,离乳47只(23雄、24雌),成活率65.9%,离乳率83.9%;18个豚鼠微卫星位点共检测到81个等位基因,平均杂合度0.5747,平均多态信息含量(PIC)0.5216。Hardy-Weinberg遗传平衡检测,14个位点符合H-W遗传平衡,3个位点显著偏离H-W遗传平衡,1个位点呈现纯合状态。结论通过剖腹产对豚鼠种群进行了净化,净化后的群体具备较理想杂合度,属高度多态性群体,符合封闭群实验动物遗传特征。Objective The guinea pig colony was decontaminated by caesarean section.Genetic testing confirms that the decontaminated colony conforms to the genetic characteristics of the closed colony.Method Female and male individuals were widely selected in different breeding units in production groups,mated at 1∶1.Cesarean section at 70 days after mating.The offspring fed by artificial milk.Breeding F1 generation by cycle mating system.DNA were extracted from 30 individuals ear tissue samples in F1 generation,18 guinea pig microsatellite markers were used to genetic testing.Result A total of 85 pups were delivered by Caesarean section,with 56 surviving and 47 weaning.Rate of surviving was 65.9%,rate of weaning was 83.9%.Eighteen microsatellites loci shared 81 alleles,the average heterozygosity was 0.5747,average polymorphism information content(PIC)was0.5216.Fourteen loci kept H-W balance.Three loci diverged significantly from the H-W balance,and 1 locus was homozygous.Conclusion The guinea pig colony was decontaminated by caesarean section and have ideal heterozygosity,belonging to highly polymorphic population,which accorded with the genetic characteristics of closed colony laboratory animals.
