摘 要:目的观察分析不同剂量碘乙酸(MIA)对于小鼠软骨和骨组织的损害,找出适合不同程度的骨关节炎小鼠模型的浓度。方法C57BL/6小鼠随机分为4组,每组左右腿互为对照注射不同剂量的碘乙酸和对照用PBS,1,2,4周后测量膝关节宽度,进行相关评分,分析病变情况。结果注射后出现不同程度病理变化。实验组关节评分明显大于对照组(P<0.05),且实验组中表现出较大剂量组(≥0.1 mg)与较小剂量组(<0.1 mg)两对组间差异。结论推荐0.1 mg为较大剂量组代表剂量,产生明显软骨组织破坏和炎症反应,0.05 mg为较小剂量组代表剂量,产生轻度软骨组织的破坏和可逆炎症反应。Objective To observe and analyze the damage on the mice cartilage and bone tissue produced by different dosages of MIA and find out the most suitable MIA dosage for building mouse osteoarthritis models.Method We choose healthy male C57 BL/6 mice randomly divided into 4 groups.The left knee was injected for MIA as treated group,the right knee was injected for PBS as control group.Knee joint width was measured 1,2 and4 weeks after the injection of MIA.Microscopic and histological scores were submitted to evaluated the damage degree of knee joint.Result We find out that different kinds of pathological change were noticed in experimental groups.The scores of the experimental groups were significantly higher than that of the control group(P<0.05).In the experimental group,there were 2 relative differences between the large dose group(≥0.1 mg)and the small dose group(<0.1 mg).Conclusion We recommended 0.1 mg as the respective dose for the large dose group,which produce obvious cartilage destruction and inflammatory,and 0.05 mg as the respective dose for the small dose group which produce mild cartilage tissue damage and reversible inflammatory reaction.
