VP2灭活疫苗与HVT-IBD载体疫苗免疫1日龄商品白羽肉鸡后保护效果比较

摘  要:将55只1日龄商品白羽肉鸡分成5组,第1~3组为免疫组(每组10只),第4组为阳性对照组(攻毒不免疫10只),第5组为阴性对照组(不免疫不攻毒15只),1~3组分别免疫市场上含有IBDV VP2蛋白的2种亚单位灭活疫苗(简称新流法)与HVT-IBD载体疫苗,21日龄使用IBDV强毒株BC6/85点眼攻毒,攻毒后4 d剖检记录法氏囊眼观病变和显微病变,以评估保护力;同时检测各组鸡只攻毒前后的VP2抗体进行分析。新流法免疫组攻毒后均有3/10的鸡出现法氏囊眼观病变,法氏囊病理评分分别为2.4和3.1,与阳性对照无明显差异,但明显高于HVT-IBD载体疫苗组(1/10法氏囊眼观病变,病理评分1.8分);攻毒前(21日龄)各免疫组VP2抗体没有显著差异,攻毒后(25日龄)新流法免疫组的抗体都有明显下降。攻毒保护及血清学结果表明:与新流法灭活疫苗相比,HVT-IBD载体疫苗1日龄免疫商品白羽肉鸡后能提供更好的保护。55 day-old commercial white broiler chickens with high titer level of maternal IBD antibody were divided into 5 groups,group1~3 were vaccination groups,group 4 was challenge positive control,group 5 was negetive control.Vaccinated group 1~3 with two kinds of inactivited vaccines containing subunit IBDV-VP2 protein(named ND-H9-VP2)and HVT-IBD vector vaccine respectively.Challenged at 21 days post vaccination using the virulent IBDV strain BC6/85 via eye drop,all the birds were killed 4 days after challenge to record the gross lesion and histological examination score of the bursa to evaluate the protective effect.Blood samples were taken before and after challenge of each group to test the VP2 antibody.3/10 birds from two ND-H9-VP2 vaccine groups had gross lesion of the bursa,the histological score of this two groups were 2.4 and 3.1 respectively,which had no significant difference with the positive control group but were significantly higher than those in the HVT-IBD vaccine group(1/10 birds had gross lesion,histological score was 1.8).There was no signifacant difference in IBD-Plus antibody among three vaccinated groups before challenge(21 days old),while after challenge(25 days old)there was a significant decrease in VP2 antibody among the two subunit inactivited vaccine groups.The vaccination challenge protection and serological results showed that compared with the two kinds of inactivited subunit IBD-VP2 vaccines,the HVT-IBD vector vaccine can provide better protection against IBD when administered to the one-day old commercial white broiler chickens.
