摘 要:作为一种嘧啶核苷,尿苷为机体多种代谢过程提供物质基础,其自身代谢和调控可能与多种代谢性疾病相关。而由脂肪细胞特异性分泌的瘦素,则通过受体作用于下丘脑神经元,在能量稳态和糖脂代谢中发挥调节作用。近年来,研究发现,尿苷可以迅速改善小鼠高脂食物喂养和衰老所致的糖耐量受损,但这一现象在瘦素缺失的小鼠中却不存在,提示尿苷对糖代谢的调节需依赖瘦素信号通路,这二者可能存在一定的协同作用。该文就脂肪细胞和进食对尿苷和瘦素的合成均起关键性调控作用进行论述,并针对二者在体温、摄食、胰岛素敏感性方面的作用及可能的协同性展开探讨。Uridine,a pyrimidine nucleoside,has a plethora of functions in the cell.Disruption of uridine homeostasis has been found associated with various metabolic diseases.Leptin,an adipokine primarily secreted by adipocytes,plays a critical role in energy homeostasis through actions at the hypothalamus.Recently,uridine administration has been found to improve glucose tolerance acutely in obese mice associated with high-fat feeding or aging but not with leptin-deficiency(ob/ob),suggesting that the impact of uridine on glucose metabolism is dependent on leptin signaling pathway.Therefore,uridine and leptin might act synergistically in glycemic control.This review summarizes the current knowledge regarding the impact of adipocytes and fasting/feeding on uridine and leptin homeostasis,and discusses their synergistic action in the regulation of core body temperature,feeding behavior,and insulin sensitivity and potential mechanism.
