摘 要:目的利用正交实验方法探究当归四逆汤四种成分芍药苷、阿魏酸、甘草酸及肉桂酸对大鼠心肌缺血再灌注损伤氧化应激机制的干预作用及最佳药物组合。方法采用结扎SD大鼠心脏冠状动脉前降支(LAD)的方法建立在体大鼠心肌缺血再灌注损伤模型。采用L16(44)正交实验设计和0.5%Evan蓝灌注心肌及1%TTC磷酸缓冲液检测血液超氧化物歧化酶(SOD)、丙二醛(MDA)水平。结果比较各因素极差发现阿魏酸、甘草酸、芍药苷、肉桂酸对大鼠心肌缺血再灌注损伤模型SOD活力影响的顺序是甘草酸>阿魏酸>芍药苷>肉桂酸;阿魏酸、甘草酸、芍药苷、肉桂酸对大鼠心肌缺血再灌注损伤模型MDA影响的顺序是肉桂酸>阿魏酸>芍药苷>甘草酸。既能降低心肌组织MDA水平,同时升高心肌SOD活力的最佳组合为阿魏酸300 mg/kg、肉桂酸200 mg/kg、甘草酸50 mg/kg。结论当归四逆汤有效成分阿魏酸300 mg/kg、肉桂酸200 mg/kg、甘草酸50 mg/kg是干预心肌再灌注损伤氧化应激的最佳组合,而芍药苷对其无明显干预作用。Objective To explore the effects of four combinations of angelica sinensis decoction including paeoniflorin,ferulic acid,glycyrrhizic acid and cinnamic acid on the oxidative stress mechanism of myocardial ischemia reperfusion injury in rats and the optimal combination by orthonal test.Methods Myocardial ischemia reperfusion injury model of rat was made by the 30 min ligation of left descending coronary artery and 120 min reperfusion in rats.Myocardial necrosis area was observed and the SOD and MDA were detected by TUNEL and perfusion of Evan blue and incubation of TTC and L16(44)orthogonal design.Results The best combination of DGSN was that ferulic acid 300 mg/kg,Cinnamic acid 200 mg/kg,Glycyrrhizic acid 50 mg/kg,which could not only reduce MDA but also increase SOD in myocardium.Conclusions A certain dose combination of ferulic acid,cinnamic acid and glycyrrhizic acid,the four effective components of angelica sinensis decoction,could not only significantly reduce the levels of MDA but also increase the activity of SOD in myocardial reperfusion injury and play a protective role,and the best cocktail combination is ferulic acid 300 mg/kg,Cinnamic acid 200 mg/kg,Glycyrrhizic acid 50 mg/kg,however the paeoniflorin has no significant effect on oxidative stress injury.
