摘  要:目的探讨西黄丸对大鼠乳腺癌癌前病变的阻断作用及对核转录因子(NF)-κB蛋白表达的影响。方法6周龄SPF级健康未育SD雌性大鼠60只,随机分为正常对照组10只,疾病模型组50只,正常对照组正常饲养,疾病模型组采用7,12-二甲基苯并蒽(DMBA)协同雌孕激素序贯建立SD大鼠乳腺癌癌前病变模型,10 w造模成功后,疾病模型组随机分为疾病模型组,他莫昔芬组,西黄丸低、中、高剂量组5组,疾病模型组予以正常饲养,他莫昔芬组(4 mg/kg)灌胃,西黄丸低、中、高剂量组(0.27、0.55、1.37 g/kg)灌胃,4 w后以水合氯醛腹腔注射麻醉各组大鼠,取大鼠乳腺组织,苏木素-伊红(HE)染色,光镜下观察病理形态学变化;免疫组化检测各组大鼠乳腺组织NF-κB蛋白表达情况。结果与疾病模型组比较,他莫昔芬组和西黄丸低、中、高剂量组乳腺病理程度较轻(P<0.01)。与正常对照组比较,疾模模型组大鼠乳腺组织NF-κB蛋白表达明显增加(P<0.05),西黄丸各剂量组可显著降低乳腺癌前病变大鼠乳腺组织NF-κB蛋白的表达水平(P<0.01,P<0.05)。结论西黄丸可阻断、逆转DMBA联合雌孕激素序贯诱导的乳腺癌癌前病变大鼠乳腺组织病理形态学变化,其机制可能与其调控乳腺组织中NF-κB蛋白的表达有关。Objective To investigate the blocking effect of Xihuang pill on precancerous lesions of breast cancer in rats and its effect on the expression of NF-kappa B protein.Methods Sixty SPF-grade SD female rats aged 6 weeks were randomly divided into normal control(n=10)and disease model groups(n=50).The normal control group was fed normally.In the disease model group,DMBA,estrogen and progesterone were sequentially used to establish the precancerous lesion model of SD rats.After 10 weeks,the disease model group was randomly divided into disease model,tamoxifen and Xihuang pill low,medium and high dose groups.The disease model group was fed normally.Tamoxifen group was given tamoxifen 4 mg/kg by gavage.The low,medium and high dose groups of Xihuang pill was given 0.27,0.55,1.37 g/kg Xihuang pill respectively by gavage.Four weeks later,rats in each group were anesthetized by intraperitoneal injection of chloral hydrate.The mammary glands of rats were taken and stained with HE.The pathomorphological changes were observed under light microscope.The expression of NF-kappa B protein in mammary glands of rats was detected by immunohistochemistry.Results Compared with disease model group,the pathological degree of mammary gland in tamoxifen group and Xihuang pill group was mild(P<0.01).Compared with normal control group,the expression of NF-kappa B protein in breast tissue of rats in model group was significantly increased(P<0.05),each dose of Xihuang pill could significantly reduce the expression of NF-kappa B protein in breast tissue of rats with precancerous lesions of breast cancer(P<0.01 and P<0.05).Conclusions Xihuang pill could block and reverse the pathological changes of mammary gland in rats with precancerous lesions induced by DMBA combined with estrogen and progesterone sequentially.Its mechanism might be related to its regulation of the expression of NF-kappa B protein in breast tissue.
