摘  要:目的探讨复方缓溃乐混悬剂(HKL)对溃疡性结肠炎(UC)大鼠紊乱代谢物的影响,并分析其治疗UC可能的作用机制。方法用2,4,6-三硝基苯磺酸(TNBS)/乙醇复合法建立UC模型;另选大鼠予以生理盐水灌肠作为正常组。按照随机数字表法将造模成功大鼠分为3组:阴性对照组、5-氨基水杨酸组(5-ASA组)和HKL组,每组12只;其中5-ASA组与HKL组大鼠分别以1 mL·200 g^-15-ASA和1 mL·100 g^-1 HKL灌胃,正常组和阴性对照组灌胃等量0.9%NaCl,均连续灌胃14 d。利用核磁共振氢谱(1H-NMR)技术结合正交偏最小二乘法-判别式分析(OPLS-DA)法筛选以上4组之间变量重要性投影值(VIP)>1的差异性代谢物。结果阴性对照组与正常组比较,阴性对照组大鼠血清中甘氨酸、乳酸、α-葡萄糖、丙酮酸和肉碱的VIP值分别为2.86,2.89,2.13,1.13和1.08。HKL组与阴性对照组比较,HKL组大鼠血清中α-葡萄糖、肉碱、牛磺酸、乳酸、极低密度脂蛋白(VLDL)的VIP值分别为5.74,1.04,5.44,7.10和2.75。与5-ASA组比较,HKL组中缬氨酸含量降低。结论HKL可调节UC大鼠所出现的代谢紊乱,其主要与调节氨基酸代谢、能量代谢及糖酵解途径有关。Objective To investigate the effect of compound Huankuile(HKL)on disordered metabolites of ulcerative colitis(UC)rats,and analyze its possible mechanism of treatment of UC.Methods The Wistar rats UC model were established by 2,4,6-trinitrobenzene sulfonic acid(TNBS)/ethanol,the other rats were randomly set as the normal group.After making the model successfully,rats were randomly divided into 3 groups:Negative control group,5-amino salicylic acid intervention group(5-ASA group)and HKL group,each group had 12 rats;among them,rats in the 5-ASA group and the HKL group were gaved with 1 mL·200 g^-15-ASA and 1 mL·100 g^-1 HKL,respectively.The normal group and the negative control group were given with an equal amount of normal saline,all were given intragastrically for 14 days.Differential metabolites with variable importance projection value(VIP)>1 between the above four groups were screened using nuclear magnetic resonance hydrogen spectroscopy(1H-NMR)technology combined with orthogonal partial least squares-discriminant analysis(OPLS-DA)method.Results Comparing the negative control group with the normal group,the VIP values of glycine,lactic acid,α-glucose,pyruvate,and carnitine in the serum of the negative control group were 2.86,2.89,2.13,1.13 and 1.08.Compared with the negative control group,the VIP values ofα-glucose,carnitine,taurine lactic acid and very low density lipoprotein(VLDL)in the serum of HKL group rats the VIP values were 5.74,1.04,5.44,7.10 and 2.75.Compared with the 5-ASA group,the valine content in the HKL group decreased.Conclusion HKL has a certain therapeutic effect in UC rats,and the mechanism may be related to regulating amino acid metabolism,energy metabolism,glycolysis.
