摘  要:目的研究白藜芦醇对非酒精性脂肪性肝病(NAFLD)小鼠血脂及炎性因子的影响。方法通过饲喂甲硫氨酸胆碱缺乏(MCD)饲料构建NAFLD小鼠模型,将NAFLD小鼠随机分为模型组和实验组,对照组饲喂正常饲料,每组15只。实验组灌胃白藜芦醇400 mg·kg^-1,每天1次,模型组和对照组灌胃等量生理盐水,连续8周。用酶联免疫吸附(ELISA)法检测小鼠血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)含量;用自动生化仪测定小鼠脂代谢水平;用苏木精-伊红(HE)染色观察小鼠肝组织病理变化;用蛋白质印迹(WB)法检测小鼠肝组织中炎性因子的表达。结果干预后,对照组、模型组和实验组小鼠肝指数为(3.54±0.29)%,(4.86±0.46)%,(4.13±0.15)%;脂肪指数为(0.43±0.11)%,(1.32±0.41)%,(0.56±0.13)%;血清中TNF-α含量为(4.43±0.42),(6.78±0.65),(5.32±0.43)pg·mL^-1;IL-6含量为(0.65±0.11),(1.65±0.22),(0.78±0.16)pg·mL^-1;血清三酰甘油(TG)含量为(0.37±0.11),(0.76±0.21),(0.54±0.12)mmol·L^-1;总胆固醇(TC)含量为(0.98±0.11),(3.64±0.21),(2.17±0.32)mmol·L^-1;游离脂肪酸(FFA)含量为(32.07±11.35),(531.65±22.34),(431.54±23.35)mmol·L^-1,差异均有统计学意义(均P<0.05)。结论白藜芦醇可通过调控NAFLD小鼠血脂及炎症因子水平而发挥改善肝组织形态的作用。Objective To investigate the effects of resveratrol on blood lipids and inflammatory factors in non-alcoholic fatty liver disease(NAFLD)mice.Methods NAFLD mice model was established by feeding methionine choline deficiency(MCD)diet,NAFLD mice were randomly divided into model group(n=15)and test group(n=15).Control group(n=15)was fed with normal diet.Test group was given resveratrol 400 mg·kg^-1,model group and control group were given saline once a day for 8 weeks.The serum levels of tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)were detected by enzyme-linked immunosorbent assay(ELISA);automatic biochemical analyzer was used to determine the level of lipid metabolism in mice;the pathological changes of liver tissues was observed by hematoxylin-eosin(HE)staining;Western blotting(WB)method was used to detect the expression of inflammatory factors in mouse liver tissues.Results After intervention,the liver index of mice in control group,model group and test group were(3.54±0.29)%,(4.86±0.46)%,(4.13±0.15)%;the fat index were(0.43±0.11)%,(1.32±0.41)%,(0.56±0.13)%;the content of TNF-αin serum were(4.43±0.42),(6.78±0.65),(5.32±0.43)pg·mL^-1;the content of IL-6 were(0.65±0.11),(1.65±0.22),(0.78±0.16)pg·mL^-1;the content of triacylglycerol(TG)in serum were(0.37±0.11)(0.76±0.21),(0.54±0.12)mmol·L^-1;the content of total cholesterol(TC)in serum were(0.98±0.11),(3.64±0.21),(2.17±0.32)mmol·L^-1,the content of free fatty acids(FFA)were(32.07±11.35),(531.65±22.34),(431.54±23.35)mmol·L^-1,the difference were all statistically significant(all P<0.05).Conclusion Resveratrol can improve the liver morphology by regulating the levels of lipid metabolism and inflammatory factors in NAFLD mice.
