摘 要:目的探讨抗帕宁汤剂对于帕金森病模型大鼠多巴胺能神经元的保护作用。方法通过MPTP偏侧纹状两点输注法构建帕金森病模型大鼠,随机将大鼠分成模型组,美多巴组,抗帕宁汤高、中、低剂量组。并将假手术组归为对照组,灌胃治疗1个月。治疗后观察各组大鼠的诱导行为变化和对纹状体单胺类神经递质、中脑酪氨酸羟化酶(TH)的影响。结果抗帕宁汤剂各剂量组经过灌胃1个月后旋转行为显著改善,且中剂量组在灌胃2周后旋转行为即见显著改善(P<0.05)。抗帕宁汤剂各剂量组大鼠损毁侧纹状体的多巴胺、二羟苯乙酸及高香草酸的含量均显著提升,其中以中剂量组增加幅度最高,差异均有统计学意义(P<0.05)。结论抗帕宁汤剂能够有效改善帕金森病模型大鼠旋转行为,并提升纹状体中多巴胺和其代谢物水平,增强TH表达,有助于保护多巴胺能神经元。采用中剂量时期效果最显著。Objective To investigate the protective effect of kangpaning Decoction on dopaminergic neurons in rats with Parkinson's disease.Methods The rats were randomly divided into model group,Madopar group and kangpaning Decoction high,medium and low dose groups.The sham operation group was divided into control group and treated by gavage for 1 month.After treatment,the changes of induced behavior and the effects on striatum monoamine neurotransmitter and midbrain tyrosine hydroxylase(TH)were observed.Results The rotation behavior of kangpaning Decoction group was significantly improved after 1 month of gavage,and the rotation behavior of middle dose group was significantly improved after 2 weeks of gavage(P<0.05).The contents of dopamine,dihydroxyphenylacetic acid and homovanillic acid in the striatum of the damaged side in each dose group of kangpaning Decoction were significantly increased,and the increase amplitude of middle dose group was the highest(P<0.05).Conclusion Kangpaning decoction can effectively improve the rotational behavior of rats with Parkinson's disease,increase the level of dopamine and its metabolites in the striatum,enhance the expression of TH,and help to protect dopaminergic neurons.The effect is most significant in the middle dose period.
