摘　　要：目的:观察脂必泰胶囊对非酒精性脂肪性肝病(NAFLD)大鼠肝组织病理、纤维化指标及炎症的影响。方法:采用高脂饲料和10%果糖诱导NAFLD大鼠模型,并随机分为模型组,脂必泰低、高剂量组(100,200 mg·kg^-1),二甲双胍组(200 mg·kg^-1),水飞蓟宾组(100 mg·kg^-1),另设正常组,造模成功后连续给药9周。肝脏称重后并计算肝脏指数,对肝组织进行HE、油红和Masson染色;检测各组大鼠血清ALT、AST、CHOL、TG、HDL-C、LDL-C,肝纤维化指标透明质酸酶(HA),层粘连蛋白(LN),IV型胶原(IV-C)和Ⅲ型前胶原(PC-Ⅲ)水平;炎症指标(IL-2、IL-6、IL-8、IL-10、IL-18、TNF-α)。结果:与模型组比较,HE、油红和Masson染色示脂必泰组肝脏病变和肝纤维化显著减轻;ALT、AST、TG、CHOL、LDL-C、肝脏脏器系数及HA、LN、IV-C、PC-Ⅲ、IL-6、IL-8、IL-10、IL-18、TNF-α明显降低,HDL-C明显升高(P<0.05)。二甲双胍组和水飞蓟宾组对NAFLD大鼠血清肝功能指标、肝纤维化指标及炎性细胞因子的改善与脂必泰一致,脂必泰高剂量组对多项指标的作用效果优于水飞蓟宾组。结论:脂必泰胶囊可降低NAFLD大鼠血脂水平,减少炎性细胞因子释放,减轻炎性反应,改善大鼠的肝脏功能和肝纤维化,对NAFLD大鼠具有良好治疗作用。OBJECTIVE To observe the effect of Zhibitai capsules on hepatic histopathology,fibrosis and inflammation in rats with nonalcoholic fatty liver disease(NAFLD).METHODS High-fat and 10%fructose diet was used to induce the rat model of NAFLD,and the rats were randomly divided into model group,the low and high dose group of Zhibitai(100 and 200 mg·kg^-1),the metformin group(200 mg·kg^-1),the silybin group(100 mg·kg^-1),and the normal control group.After the establishment of the models,the rats were dosed continuously for 9 weeks.The liver was weighed and the liver index was calculated,and the liver tissues were stained with HE,oil red and Masson;the serum levels of ALT,AST,CHOL,TG,HDL-C,LDL-C;liver fibrosis indexes such as HA(hyaluronidase),laminin(LN),type IV collagen(IV-C)and type III collagen(PC-Ⅲ);and inflammatory indexes(IL-2,IL-6,IL-8,IL-10,IL-18 and TNF-α)were determined.RESULTS Compared with model group,HE,oil red and Masson staining showed significant alleviation of liver lesions and fibrosis in Zhibitai groups;ALT,AST,TG,CHOL,LDL-C,liver viscera coefficient,HA,LN,IV-C,PC-Ⅲ,IL-6,IL-8,IL-10,IL-18 and TNF-αwere significantly decreased;HDL-C was significantly increased(P<0.05).Metformin and silybin had the similar effects on liver function,liver fibrosis,and inflammatory cytokines,and the effects of high dose of Zhibitai were better than those of silybin.CONCLUSION Zhibitai capsules could modulate blood lipid,reduce the release of inflammatory cytokines,alleviate inflammation,improve liver function and fibrosis,and has a good therapeutic effect on NAFLD rats.
