摘  要:[目的]比较液体保存、液氮保存与冷冻保存半月板的形态与生物力学特点,为临床半月板同种异体移植奠定基础。[方法]获取成年健康猪的内侧半月板60个,随机分成3组,分别采用液体保存、液氮保存和冷冻保存,于2周、4周和8周时检测保存半月板的形态学和杨氏模量改变。[结果]随保存时间延长,半月板形态退变逐步加重,液体保存和液氮保存半月板组的形态优于冷冻保存。随时间推移,三组半月板O染色和甲苯胺蓝染色IOD值显著降低(P<0.05),液体保存组和液氮保存组半月板的番红O染色和甲苯胺蓝染色IOD值显著高于冷冻保存组(P<0.05)。生物力学检测方面,三组的杨氏模量显著下降(P<0.05);液体保存和液氮保存组半月板杨氏模量显著高于冷冻保存组(P<0.05)。[结论]液体保存和液氮保存半月板在形态和力学特征方面均明显优于冷冻保存半月板。[Objective] To compare morphological and biomechanical characteristics of menisci preserved by three methods to provide a reference for meniscus allograft storage in clinical setting. [Methods] A total of 60 medial meniscus were harvested from adult healthy pigs, and were randomly divided into 3 groups with 20 menisci in each group. Of them, 20 menisci were preserved with fluid technique(the FT group), 20 were by cryopreservation with liquid nitrogen(-196℃)(the LN group),while the remaining 20 menisci were stored by fresh-frozen preservation(-80 ℃)(FFP group). Micromorphology and biomechanical assay were conducted respectively at week 2, 4 and 8 after preservation. [Results] The menisci progressively degenerated in histological observations with HE staining, Safrin O and toluidine blue staining over time in all the 3 groups(P<0.05),which proved less significant in the FT and LN group than the FFP group at any time point after preservation(P<0.05). The IOD value of Safrin O and toluidine blue staining declined significantly as time went in all the 3 group(P<0.05), which was significantly higher in the FT and LN group than the FFP group(P<0.05). In term of biomechanical test, the Young’s modulus significantly decreased over time in the 3 groups(P<0.05), whereas which was higher in the FT and LN group than the FFP group(P<0.05). [Conclusion] The menisci preserved by fluid technique and cryopreservation are superior to the fresh-frozen preservation in morphological and biomechanical features.
