摘 要:目的优化翅茎白粉藤总黄酮的超声提取工艺并考察其体外抗氧化活性。方法在单因素试验的基础上,通过正交试验,分析乙醇体积分数、料液比、提取温度、提取时间对翅茎白粉藤总黄酮得率的影响,并优化提取工艺;以维生素C为阳性对照评价其清除DPPH+·和ABTS+·的能力。结果超声提取翅茎白粉藤总黄酮最佳工艺条件为乙醇体积分数75%、料液比1∶25(g/mL)、提取温度40℃、提取时间40 min,总黄酮平均得率为2.38%。总黄酮对DPPH+·和ABTS+·清除率分别可高达 95.59%(总黄酮质量浓度250 μg·mL^-1)和 98.11%(总黄酮质量浓度1000 μg·mL^-1),有较强的抗氧化能力。结论该方法可为翅茎白粉藤总黄酮的提取研究和应用提供一定参考依据。Objective To optimize the ultrasonic-assisted extraction of total flavonoids from Cissus hexangularis and to determine its antioxidant activities.Methods The effects of ethanol concentration,material-liquid ratio,extraction temperature and extraction time on the extraction yield of total flavonoids were analyzed by the single factor test.The extraction process was optimized by orthogonal experiment.The in vitro antioxidant activities of the total flavonoids were determined by clear DPPH+·and ABTS+·,with vitamin C as the positive control.Results The optimal ultrasonic-assisted extraction conditions were 75% ethanol,material-liquid ratio at 1:25 (g/mL),extraction temperature at 40℃ and extraction for 40 min.The average yield of total flavonoids was 2.38%.The total flavonoids of Cissus hexangularis scavenging rate of DPPH+·and ABTS+ ·were respectively 95.59% (total flavonoids concentration at 250 μg·mL^-1),and 98.11% (total flavonoids concentration at 1000 μg·mL^-1),exhibiting strong antioxidant activities.Conclusion This study provides a basis for further research and development of total flavonoids from Cissus hexangularis.
