摘 要:目的观察131I-牛血清白蛋白(BSA)-聚己内酯(PCL)-索拉非尼(sorafenib)在人甲状腺未分化肿瘤细胞8305C荷瘤裸鼠体内的靶向性分布及其抑瘤作用。方法制备并鉴定BSA-PCL-sorafenib、131I-BSA-PCL-sorafenib脂质体。12只裸鼠接种人甲状腺未分化肿瘤细胞8305C建立模型,造模成功后分别尾静脉注射生理盐水(对照组)、131I、BSA-PCL-sorafenib和131I-BSA-PCL-sorafenib,通过SPECT/CT显像观察注射后不同时间荷瘤裸鼠体内的放射性分布,比较各组荷瘤裸鼠的体质量及肿瘤质量和体积变化。结果温育60 min后,131I-BSA-PCL-sorafenib组细胞荧光强度高于对照组。注射22 d后,131I-BSA-PCL-sorafenib组的肿瘤质量和体积最小。结论131I-BSA-PCL-sorafenib可有效抑制8305C细胞造模诱发的肿瘤生长,在未分化型甲状腺癌的治疗和预后评估中有良好的应用前景。OBJECTIVE To observe the radioactivity distribution and antitumor efficacy of 131I-bovine serum albumin(BSA)-polycaprolactone(PCL)-sorafenib in human anaplastic thyroid carcinoma cells 8305C-bearing nude mouse model.METHODS Prepared and identified BSA-PCL-sorafenib,131I-BSA-PCL-sorafenib liposomes.Twelve nude mice were inoculated with human anaplastic thyroid carcinoma cells 8305C to establish model.After successful modeling,normal saline(control group),131I,BSA-PCL-sorafenib,and 131I-BSA-PCL-sorafenib were injected into the tail vein.The radioactivity distribution in tumor-bearing nude mice at different times after injection was observed by SPECT/CT imaging,and the body weight and tumor mass and volume changes of tumor-bearing nude mice in each group were compared.RESULTS After incubation for 60 min,the fluorescence intensity in 131I-BSA-PCL-sorafenib group was higher than that in control group.After injection for 22 d,the weight and volume of tumor in 131I-BSA-PCL-sorafenib group was the smallest one in four groups.CONCLUSION 131I-BSA-PCL-sorafenib can effectively inhibit the tumor growth of 8350C induced-mouse tumor model,which gives a good suggestion for the treatment and prognosis evaluation of anaplastic thyroid carcinoma.
