摘 要:目的:探讨不同浓度姜黄素对S180肉瘤细胞的增殖及体外血管生成拟态的影响及其作用机制.方法:用不同浓度姜黄素(5~20μmol/L)处理S180肉瘤细胞,MT法检测S180肉瘤细胞增殖活性;在Matrigel上接种S180肉瘤细胞,观察S180肉瘤细胞是否能生成血管拟态以及其结构特点;并通过体外血管拟态抑制实验检测姜黄素对S180肉瘤细胞管道形成能力的影响;采用Wstern blot法检测血管生成拟态中基质金属蛋白酶2(matrixmetaclo proteinase-2,MMP-2)抗体、MMP-9蛋白分子表达。结果:姜黄素对肉瘤细胞的增殖具有抑制作用,与对照组比较差异有统计学意义(P<0.05),且浓度越高,时间越长,抑制作用越明显,具有剂量效应和时间效应关系。S180肉瘤细胞经培养能够形成血管拟态结构,并且能和内皮细胞相互作用形成马赛克样的血管网状结构;姜黄素对S180肉瘤细胞体外血管拟态的形成具有抑制作用,与对照组比较差异有统计学意义(P<0.05),具有剂量效应关系,其管状结构的数量与姜黄素的浓度呈负相关(r=-0.672,P<0.05),并且能够降低血管生成拟态中MMP-2、MMP-9蛋白分子的表达。结论:姜黄素可能通过下调MMP-2、MMP-9蛋白分子表达来实现对S180肉瘤细胞的增殖抑制作用及抑制S180肉瘤细胞体外血管拟态结构形成。Objective: To discuss the effects of different concentrations of curcumin on the proliferation of S180 sarcoma cells and vasculogenic mimicry in vitro, and its mechanism. Methods: After dealing S180 sarcoma cells with different concentrations of curcumin(5 to 20 μmol/L), MTT method was used to detect the proliferative activity of S180 sarcoma cells;S180 sarcoma cells were inoculated on Matrigel to observe whether S180 sarcoma cells could produce vasculogenic mimicry and its structural features;the effects of curcumin on pipeline forming capacity were detected via the inhibitory experiments of vascular mimicry in vitro;and molecular expressions of MMP-2 antibody and MMP-9 protein in vasculogenic mimicry were measured using Western blot method. Results:Curcumin owned the inhibitory effects on the proliferation of S180 sarcoma cells, the difference showed statistical meaning compared with the control group(P<0.05), the inhibitory effects were more notable when the concentrations became stronger and the time were longer, in dose-effects relationship and time-effects relationship. S180 sarcoma cells could form the structure of vascular mimicry, and interact with endothelial cells to form a mosaic like vascular network;curcumin could inhibit the formation of vascular mimicry in vitro of S180 sarcoma cells, the difference showed statistical meaning compared with the control group(P<0.05), in dose-effect relationship, the number of tubular structure was negatively related to the concentrations of curcumin(r=-0.672, P<0.05), and it could lower the expressions of MMP-2 and MMP-9. Conclusion: Curcumin could realize the aim of inhibiting proliferation on S180 sarcoma cells and vasculogenic mimicry in vitro through down regulating the expressions of MMP-2 and MMP-9.
