摘 要:该研究旨在建立基于线粒体COⅠ基因的鸭源性成分荧光定量PCR检测方法。以猪、牛、羊、兔、鸡、鸭、鹅、鸽和鹌鹑9种动物线粒体DNA COⅠ基因为靶基因,寻找差异位点,设计筛选出鸭特异性引物,以9种动物肌肉DNA为模板,进行常规PCR和荧光定量PCR扩增,并将鸭肉DNA模板浓度按10倍、102倍、103倍、104倍、105倍、106倍、107倍和108倍等8个梯度进行稀释,检测该方法灵敏度。所设计筛选的引物能够有效地扩增出畜禽肉中鸭源性成分,特异性较强,灵敏度较高。所建立的方法能够快速准确地鉴别出畜禽肉中鸭源性成分。In order to establish a fluorogenic quantitative PCR method which could identify the components of duck origin,Mitochondrial DNA COⅠgene of pigs,cows,sheep,rabbits,chickens,ducks,geese,pigeons and quails were used as target sites and the duck specific primers were designed and screened.Nine animals'DNA were used as templates for conventional PCR and fluorescence quantitative PCR,and the duck DNA template was diluted in eight gradients,from 10 times to 108 times,for detecting the sensitivity.It showed that the design screening primers could detect the duck origin effectively,which had a quite specificity and high sensitivity.The established method could detect the duck origin in livestock and poultry meat quickly and accurately.
