苏木乙酸乙酯提取物对大鼠的毒理研究

摘　　要：目的评价苏木乙酸乙酯提取物给药26周对大鼠的安全性。方法分别以0.09、0.38、1.00 g/kg的剂量,每日1次灌胃雄性和雌性大鼠并于给药13周末、26周末处死大鼠。在给药13周、26周和恢复4周后对大鼠进行系统检查,采集指标包括血液学、血液生物化学和组织形态学。结果一般情况下,食物摄入、体质量及组织形态学检查中未观察到治疗相关的不良反应。在治疗期间,苏木乙酸乙酯提取物对部分血液学和生物化学指标以及相关器官重量进行了修改,但所有变化均在正常范围内,并在4周停止治疗后均恢复到基线水平。结论在该研究的26周给药期内,以1.00 g/kg剂量(相当于临床剂量的55.5倍)给药的大鼠对苏木乙酸乙酯提取物耐受性良好,无明显毒性。Objective To evaluate the safety of Caesalpinia sappan L.extract administered for 26 weeks in Sprague Dawley rats.Methods C.sappan L.extract was administered to male and female rats by oncedaily gavage at doses of 0.09,0.38,or 1.00 g/kg.The rats were sacrificed after intragastric administration lasting 13 or 26 weeks.A subset of rats in each group was allowed to recover for 4 weeks after 26-week administration.Systematic examinations including hematology,blood biochemistry,and histomorphology were performed at the end of 13 or 26 weeks of administration or following 4 weeks of recovery.Results No treatment-related adverse effects were observed on general condition,food intake,body weight,or histomorphological examinations.During the treatment period,some hematology and biochemistry values and relative organ weights were modified by C.sappan L.,but all changes were within the normal ranges and all returned to baseline after discontinuing treatment at 4 weeks.Conclusion The administration of ethyl acetate extract of C.sappan L.at doses up to 1.00 g/kg(equivalent to 55.5 times the clinical dose)was well tolerated without significant toxicity within the 26-week administration period in this study.
