摘 要:目的比较中药葛根、葛花及其总黄酮对酒精诱导的急性肝损伤大鼠的防治作用差异及其作用机制。方法采用白酒灌胃法建立急性酒精性肝病模型。将SPF级SD大鼠(雄性,70只),随机分为7组,每组10只,包括:正常组、模型对照组、联苯双酯阳性药对照组、葛根组、葛花组、葛根总黄酮组和葛花总黄酮组。连续造模并治疗15 d后采集大鼠血清、肝脏组织。自动生化分析仪检测谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)、尿素氮(blood urea nitrogen,BUN)、甘油三酯(triglyceride,TG)、总胆固醇(total cholesterol,TC)、高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL-C)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)。肝组织采用苏木精-伊红染色法(hematoxylin-eosin staining,HE)染色,显微观察肝组织病理变化。结果与模型组相比,各治疗组均可以显著降低大鼠血清尿素氮和血清甘油三酯水平,而血清总胆固醇,高、低密度脂蛋白胆固醇各组间无明显差异,仅葛根组可以显著降低血清总胆固醇和高、低密度值蛋白胆固醇水平。结论过量白酒可引起肝肾损伤,葛根、葛花在治疗酒精性肝损伤方面可以互补;葛根、葛花总黄酮为解酒活性成分,但解酒效果不如总提物。Objective To compare the prevention and treatment of acute liver disease induced by alcohol in rats with Radix Puerariae lobatae,Puerariae lobatae Flos and their total flavonoids.Methods The acute alcoholic liver disease model was established by ethanol gavage.70 male SPF grade Sprague Dawley rats were randomly divided into 7 groups,10 for each group,including:normal group,model control group,bifendate positive group,Radix Puerariae lobatae,Puerariae lobatae Flos group,Radix Puerariae lobatae total flavone group and Puerariae lobatae Flos total flavone group.Serum and liver tissue were collected after 15 days.Automatic biochemical analyzer was used for detecting serum alanine aminotransferase,aspartate aminotransferase,blood urea nitrogen,triglyceride,Total cholesterol,high density lipoprotein cholesterol and low density lipoprotein cholesterol.Hematoxylin-eosin staining method was used to observe the pathological changes of liver tissue.Results Compared with the model group,all the treatment groups could significantly reduce the serum urea nitrogen and serum triglyceride levels,while there was no significant difference between the groups of total cholesterol,high and low density lipoprotein cholesterol,and only the Radix Puerariae lobatae group could significantly reduce level of total cholesterol and high and low density value protein cholesterol.Conclusion Excessive white spirit taken can cause liver and kidney injury.The flavonoids of Radix Puerariae lobatae and Puerariae lobatae Flos were the active components,but the effect was not as good as the total extracts.
