摘  要:目的构建桃仁-红花药对成分靶点网络与蛋白作用网络,探究桃仁-红花药对"异病同治"慢性稳定型心绞痛和失眠的作用靶标、信号通路。从分子生物学、系统生物学、生物信息学等角度,分析活血化瘀药对桃仁-红花"异病同治"慢性稳定型心绞痛和失眠的作用机制。方法采用网络药理学方法。结合文献资料,应用中药系统药理学分析平台(TCMSP)对桃仁-红花药对活性成分及作用靶点进行筛选(口服利用度OB≥30%,类药性DL≥0.18)。结合TTD、DrugBank、OMIM数据库获取慢性稳定型心绞痛和失眠疾病相关靶点。采用Cytoscape软件绘制药对-成分-靶点网络。利用STRING数据库,构建出蛋白-蛋白相互作用PPI网络模型,并利用Cytoscape软件进行优化;利用DAVID数据平台,对靶标本体GO生物过程和KEGG代谢通路进行富集分析。结果经筛选,桃仁-红花药对与慢性稳定型心绞痛57个靶点交集,失眠29个靶点交集。GO生物过程结果显示治疗慢性稳定型心绞痛和失眠途径为一氧化氮生物合成过程正调控、炎症反应过程、G蛋白偶联乙酰胆碱受体信号通路等。KEGG通路富集分析,结果显示共同涉及调节TNF信号通路、Toll样受体信号通路、神经活性配体-受体相互作用等。表明桃仁-红花药对可能通过免疫炎症反应、神经阻断过程、基因表达过程等发挥对慢性稳定型心绞痛与失眠的共同治疗作用。结论桃仁-红花药对是通过多成分、多靶点、多种信号通路来发挥抗稳定性心绞痛及失眠的作用,为深入阐释桃仁-红花药对"异病同治"的理念及"活血化瘀"法的临床应用提供依据。Objective Construction of Peach kernel and Carthamus tinctorious couplet medicine interaction network on component targets and protein,and explore targets and signaling pathways of Peach kernel and Carthamus tinctorious couplet on stable angina pectoris and insomnia.From the perspectives of molecularbiology,systems biology and bioinformatics,analysis of drugs for promoting blood circulation and removing blood stasis the mechanism of stable angina pectorisand insomnia.Methods The network pharmacology method.Application TCMSP data base to screening of active components and targets of Peach kernel and Carthamus tinctorious couplet(OB≥30%,DL≥0.18).Targets related to stable angina pectoris and insomnia can be obtained by TTD,DrugBank,OMIM database.Drawing drug pair-componenttarget network with Cytoscape software.Using STRING database,construction of PPI network model of protein-protein interaction;Using DAVID database,enrichment and analysis of GO biological processes and KEGG metabolic pathways in target specimens.Results After screening,Peach kernel and Carthamus tinctorious couplet intersects 57 targets with stable angina pectoris,and 29 targets with insomnia.GO biological process results,treatment of stable angina pectoris and insomnia were positive regulation of nitric oxide biosynthetic process,inflammatory response,G-protein coupled acetylcholine receptor activity and so on.KEGG metabolic pathways results were involved TNF signaling pathway,Toll-like receptor signaling pathway,neuroactive ligand-receptor interaction and so on.It was suggested that Peach kernel and Carthamus tinctorious couplet may play a co-therapeutic role in stable angina pectoris and insomnia through immunoinflammatory reaction,nerve blockade and gene expression.Conclusion Peach kernel and Carthamus tinctorious couplet medicine plays an anti-stable role in stable angina pectoris and insomnia through multicomponent,multi-target and multi-signal pathway,to provide a basis for further elucidating the concept of"treating different diseases w
