摘 要:目的采用网络药理学的方法,对宣痹汤治疗类风湿关节炎的作用机制进行研究探讨。方法通过在TCMSP平台对宣痹汤进行活性成分筛选、在DrugBank数据库进行靶点预测,应用Cytoscape 3.7.1软件构建成分-靶点网络;在TTD及DisGenet数据库中进行检索,获取疾病靶点;构建韦恩图,取疾病与药物靶点交集,得到宣痹汤治疗RA的作用靶点;并进行GO功能注释和KEGG通路富集分析;构建宣痹汤中药-成分-靶点-信号通路网络,使用network analyzer进行网络特征分析。结果研究显示共挖掘到宣痹汤中59种成分及271个潜在靶点,RA疾病1838个相关靶点,得到宣痹汤-疾病共同靶点29个,共同作用靶点主要富集于76个生物过程和51条信号通路上。结论宣痹汤可能是通过调节MAPK1、JUN、TNF、IFNG等靶点,调控TNF信号通路、T细胞受体信号通路、IL-17信号通路、Th17细胞分化、破骨细胞分化等通路,从而抑制炎症反应、调节免疫功能来治疗类风湿关节炎。Objective To study the mechanism of Xuanbi Decoction in the treatment of rheumatoid arthritis(RA) by means of network pharmacology.Methods The active ingredients of Xuanbi Decoction were screened on TCMSP platform.The target was predicted in DrugBank database,and the component-target network was constructed with Cytoscape 3.7.1 software.The disease targets were retrieved in TTD and DisGenet databases and the Wein map was constructed to intersect the disease and drug targets,and the targets of treatment of Xuanbi Decoction was obtained.GO function annotation and KEGG pathway enrichment analysis were carried out.The Xuanbi Decoction traditional Chinese medicine-components-targets-signaling pathway network was set up and the network analyzer was used for for network characteristics analysis.Results A total of 59 components and 271 potential targets of Xuanbi Decoction were found,1838 related targets of RA disease were found,and 29 common targets of Xuanbi Decoction and disease were obtained.The targets of interaction were mainly concentrated in 76 biological processes and 51 signaling pathways.Conclusion Xuanbi Decoction may regulate TNF signaling pathway,osteoclast differentiation,T cell receptor signaling pathway,IL-17 signaling pathway and Th17 cell differentiation by regulating MAPK1,JUN,TNF and IFNG,so as to inhibit inflammatory response and regulate immune function in the treatment of rheumatoid arthritis.
