摘 要:目的运用响应曲面法的Box-Behnken设计,研究玛咖多糖的最佳提取条件。方法苯酚-硫酸法作为多糖的测定方法。选择原料粒度、提取温度、酒精浓度作为优化因素,研究各因素的不同水平对玛咖多糖提取效率的影响。结果通过响应曲面分析得到玛咖多糖的最优提取条件为:原料粒度为0.15~0.18 mm,提取温度为10℃,酒精浓度为0%。结论所建立的回归模型有效,玛咖多糖最优理论提取效率可达13.11 mg/g,实际提取效率为12.84 mg/g,与理论值的相对误差为-2.1%。Objective Box Behnken method of Response Surface Methodology(RSM)was used to explore the optimal extraction conditions of maca polysaccharide.Methods Phenol-sulfuric acid method was used for the determination of maca polysaccharide.In RSM analysis,the effects of raw material size,extraction temperature and alcohol concentration on the extraction efficiency of maca polysaccharide were investigated.Results The optimal extraction conditions of maca polysaccharide were as follows:0.15-0.18 mm of maca granularity,10℃of extraction temperature,0%of alcohol.Conclusion The regression model established is valid,the practical extraction efficiency for maca polysaccharide was 12.84 mg/g in the experiment,which was consistent with the theoretical value(13.11 mg/g),and the relative deviation was-2.1%.
