中医穴位敷贴戒烟对人体尿液代谢组学的影响

摘　　要：目的了解使用中医穴位敷贴戒烟30天后尿液代谢组学差异,探索相关的代谢通路。方法采集14名成功戒烟者戒烟前、后尿液标本,利用液相色谱质谱联用(LC-MS)非靶向代谢组分析尿液代谢物,采用主成分分析(PCA)、正交偏最小二乘法判别分析(OPLS-DA)等多元统计分析方法,对戒烟前、后尿液代谢物进行分析,筛选出潜在生物标志物;借助差异代谢物的KEGG ID进行通路富集分析,获得代谢通路富集结果。结果与戒烟前相比,戒烟后试验参与者尿液代谢物可明显区分,差异有统计学意义(P值均<0.05),其中维生素的消化和吸收代谢、尼古丁成瘾代谢、色氨酸代谢和癌症中的胆碱代谢通路,戒烟前后富集程度具有统计学意义(P值均<0.05)。结论中医穴位敷贴可能通过调节维生素消化和吸收代谢、尼古丁成瘾代谢和色氨酸代谢等代谢通路,缓解戒烟后的戒断反应,恢复机体代谢平衡,达到戒烟目的。Objective To investigate the differential metabolites in urine metabolomics of 30 dsmoking cessation with traditional acupuncture paste application;to explore the relevant metabolic pathways.Methods A total of 14 subjects quitted smoking successfully were enrolled in this study,whose urine samples were collected before and after smoking cessation.The urine metabonomics were analyzed by liquid chromatography mass spectrometry(LC-MS),and the quantitative data were further processed by principal component analysis(PCA)and orthogonality PLS-DA(OPLS-DA)to identify potential biomarkers.In addition,KEGG ID analysis was used for pathway analysis to acquire metabolic pathway enrichment results.Results Compared with indexes before smoking cessation,the urine metabolites of the participants were significantly different after quitting smoking,all differences were statistically significant(all P<0.05).The vitamins digestion and absorption metabolism,nicotine addiction metabolism,tryptophan metabolism and choline metabolism pathway in cancer were statistically significant before and after smoking cessation(all P<0.05).Conclusion The Chinese traditional medicine acupuncture paste might alleviate the withdrawal reaction of smoking cessation,restore the metabolic balance of the body by regulating the metabolism of vitamin digestion and absorption,nicotine addiction and Tryptophan to achieve the purpose of smoking cessation.
