摘 要:目的探讨莱菔硫烷(sulforaphane,SFN)对AlCl 3诱导人神经母细胞瘤细胞(SH-SY5Y)细胞毒性的保护作用及其机制。方法体外培养SH-SY5Y,采用不同浓度AlCl 3染毒24 h,或采用经不同浓度SFN预处理SH-SY5Y细胞24 h后进行AlCl 3染毒24 h,采用流式细胞仪检测SH-SY5Y细胞周期和凋亡情况;采用Hochest染色法检测SH-SY5Y细胞凋亡情况;采用western blot法检测SH-SY5Y细胞周期相关蛋白(Cyclline D1、P27、P21)和凋亡相关蛋白(Cyto C、Caspase 9、Caspase3)表达水平。结果AlCl 3可引起细胞周期阻滞,SFN对其周期阻滞具有抑制作用。随着AlCl 3浓度的增加,SH-SY5Y细胞的凋亡率增高,而SFN可以降低细胞凋亡率,染毒后,可见细胞核形态改变,凋亡现象明显。AlCl 3诱导SH-SY5Y细胞周期相关蛋白和凋亡相关蛋白的表达,SFN对其相关蛋白的表达均具有抑制作用。结论铝可以阻滞细胞周期、使细胞凋亡率增高,SFN对铝诱导上述的改变具有抑制作用。Objective To investigate the protective effect of sulforaphane(SFN)on the cytotoxicity of human neuroblastoma cells(SH-SY5Y)induced by AlCl 3 and the mechanism.Methods SH-SY5Y were cultured in vitro.The SH-SY5Y cells were infected with AlCl 3 of different concentrations for 24 hours or pretreated with SFN of different concentrations for 24 hours and then infected with AlCl 3 for 24 hours.The cell cycle and apoptosis of SH-SY5Y cells were detected by flow cytometry.The apoptosis of SH-SY5Y cells was detected by Hochest staining.Western blot was used to detect the expressions of cycle-related proteins(Cyclline D1,P27,P21)and apoptosis-related proteins(Cyto C,Caspase 9,Caspase3)in SH-SY5Y cells.Results AlCl 3 could cause cell cycle arrest;SFN had an inhibitory effect on cell cycle arrest.With the increase of AlCl 3 concentration,the apoptosis rate of SH-SY5Y cells increased,while SFN could decrease the apoptosis rate.After being infected,the morphology of the cell nucleus changed and apoptosis was observed.AlCl 3 induced the expressions of cell cycle-related proteins and apoptosis-related proteins in SH-SY5Y cells,and SFN inhibited the expression of these proteins.Conclusion Aluminum can block cell cycle and increase apoptosis rate of cells,and SFN can inhibit these changes induced by aluminum.
