摘  要:目的研究穿心莲内酯(andrographolide,AND)对人胶质瘤(Glioma)细胞U87-MG生长和凋亡的影响。方法体外培养人胶质瘤细胞U87-MG细胞株,采用MTT法检测AND对U87-MG细胞活力的影响;Hoechst 33342染色实验观察AND对U87-MG细胞凋亡的影响;JC-1染色实验观察AND处理后细胞线粒体膜电位(mitochondrial membrane potential,MMP)变化;Western blot法检测Bax、Bcl-2、JAK2、p-JAK2、STAT3、p-STAT3蛋白表达水平。结果AND能显著降低U87-MG细胞活力,且具有浓度依赖性和时间依赖性;Hoechst 33342染色实验显示,AND作用于U87-MG细胞24 h后,细胞核明显浓缩,细胞凋亡数量增加;JC-1染色实验显示,随着AND浓度增加,绿色荧光增强,表明细胞线粒体膜电位降低;Western blot法检测结果显示,AND可上调U87-MG细胞Bax蛋白表达,同时下调Bcl-2、JAK2、p-JAK2、STAT3、p-STAT3蛋白表达。结论AND能抑制胶质瘤细胞U87-MG的生长并诱导其凋亡,其分子机制与JAK2/STAT3信号通路有关。Objective To study the effect of andrographolide(AND)on the growth and apoptosis of human glioma cell U87-MG.Methods The U87-MG glioma cell lines of human were cultured in vitro,and MTT assay was used to detect the effect of AND on the activity of U87-MG cells.Hoechst 33342 staining was conducted to observe the effect of AND on the apoptosis of U87-MG cells and JC-1 staining to observe the changes of cells mitochondrial membrane potential(MMP)after AND treatment.The protein expressions of Bax,Bcl-2,JAK2,p-JAK2,STAT3 and p-STAT3 were detected by Western blot.Results AND could significantly reduce the activity of U87-MG cells in a concentration-dependent and time-dependent manner.Hoechst 33342 staining showed that the nuclei of U87-MG cells were significantly concentrated and the number of apoptosis increased after AND being applied to U87-MG cells for 24 hours.JC-1 staining revealed that with the increase of AND concentration,green fluorescence was enhanced,indicating MMP decreasing.Western blot detection showed that AND could up-regulate the Bax protein expression in U87-MG cells,and down-regulate the protein expressions of Bcl-2,JAK2,p-Jak2,STAT3 and p-Stat3.Conclusion AND can inhibit the growth of U87-MG glioma cells and induce their apoptosis,and its molecular mechanism is related to JAK2/STAT3 signaling pathway.
