摘  要:目的探讨鸡骨草提取物在体内对SGC-7901胃癌细胞、BEL-7404肝癌细胞、MCF-7乳腺癌细胞裸鼠移植瘤生长的影响。方法用95%乙醇对鸡骨草进行提取,并分别用乙酸乙酯、正丁醇和水对其依次进行萃取,选用SGC-7901、BEL-7404、MCF-7细胞在裸鼠腋下接种,创建裸鼠皮下移植瘤模型。当肿瘤长到50 mm 3时,采用随机分配的方法分为模型组,鸡骨草乙酸乙酯、正丁醇、水提取物的低、中、高剂量组,模型组给予生理盐水,实验组每天灌胃剂量为1 g/kg、2 g/kg、4 g/kg的鸡骨草乙酸乙酯、正丁醇、水提取物,连续给药21 d。取出瘤体,用公式计算肿瘤体积及肿瘤抑制率,HE染色法观察组织的细胞病变情况,免疫组化检测乙酸乙酯提取物组肿瘤组织中Bax和Bcl-2两种蛋白的表达。结果鸡骨草各提取物体内均具有抑制肿瘤生长的作用,以鸡骨草乙酸乙酯提取物组抑瘤作用最强,正丁醇提取物组抑瘤效果次之,水提取物组最差,HE染色结果显示,与模型组相比,鸡骨草各提取物组肿瘤细胞出现凋亡形态改变。免疫组化结果显示,鸡骨草乙酸乙酯提取物组能下调肿瘤组织中Bcl-2蛋白的表达同时上调Bax蛋白的表达。结论鸡骨草乙酸乙酯、正丁醇、水提取物对肿瘤细胞裸鼠移植瘤有抑制作用,并能够诱导细胞凋亡,乙酸乙酯提取物可上调Bax蛋白和下调Bcl-2蛋白的表达。Objective To investigate the effect of Abruscantonensis extract in vivo on the growth of xenograft tumors caused by SGC-7901 gastric cancer cells,BEL-7404 hepatoma cells and MCF-7 breast cancer cells in nude mice.Methods Abruscantoniensis was extracted by 95%ethanol;then the ethanol extract was extracted with ethyl acetate,n-butanol alcohol and water sequentially.The SGC-7901,BEL-7404 and MCF-7 cells were inoculated under the armpit of nude mice to create subcutaneous xenograft tumor models in nude mice.When the cancer grew up to 50 mm 3,the nude mice were randomly divided into the model group,the low,medium and high dose groups of ethyl acetate,n-butanol alcohol and water extract from Abruscantoniensis.The model group was given normal saline,while the experimental groups were given 1 g/kg,2 g/kg and 4 g/kg doses of ethyl acetate,n-butanol alcohol and water extract from Abruscantoniensis by gavage daily for 21 days.The tumors were removed,and the tumor volume and the tumor inhibition rate were calculated by formula.The cytopathic condition of the tissue was observed by HE staining.Immunohistochemistry was used to detect the protein expressions of Bax and Bcl-2 in the tumor tissues of the ethyl acetate extract group.Results All the extracts from Abruscantoniensis had the inhibitory effect on tumor growth in vivo.The ethyl acetate extract group had the strongest inhibition effect,followed by the n-butanol alcohol extract group and the water extract group.HE staining results showed that,in comparison with the model group,all the Abruscantoniensis extract groups had morphological changes of apoptosis of tumor cells.Immunohistochemistry showed that the ethyl acetate extract of Abruscantonensis could down-regulate the expression of Bcl-2 protein and up-regulate the expression of Bax protein at the same time.Conclusion The ethyl acetate,n-butanol alcohol and water extract from Abruscantoniensis can inhibit the growth of SGC-7901,BEL-7404,MCF-7 cancer cells in xenograft tumors of nude mice.The ethyl acetate extract of Ab
