摘　　要：目的研究流产布鲁菌变异株S2308△rfbE感染对小鼠髓源性树突状细胞(BMDC)成熟和免疫应答功能的影响。方法采用流式细胞术检测S2308△rfbE感染小鼠BMDC后主要组织相容性抗原复合体Ⅰ(MHCⅠ)、MHCⅡ、CD40、CD86的表达变化,ELISA检测BMDC的肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)和IL-10的分泌量。结果S2308△rfbE感染BMDC后,细胞表达MHCⅠ、MHCⅡ、CD40、CD86明显增强,TNF-α、IL-6和IL-10分泌量升高。结论布鲁菌变异株S2308△rfbE促进BMDC成熟和细胞因子分泌。Objective To investigate the effect of Brucella abortus mutant strain S2308△rfbE on the maturation and immune response of mouse bone marrow-derived dendritic cells(BMDCs).Methods The expression of major histocompatibility complexⅠ(MHCⅠ),MHCⅡ,CD40,CD86 were detected by flow cytometry;the secretion of cytokines tumor necrosis factorα(TNF-α),interleukin 6(IL-6),IL-10 was detected by ELISA in S2308△rfbE-infected mouse BMDCs.Results The number of cells expressing MHCⅠ,MHCⅡ,CD40,CD86 and the secretion of cytokines TNF-α,IL-6 and IL-10 significantly increased in BMDCs infected with S2308△rfbE as compared with control groups.Conclusion S2308△rfbE promotes the maturation and enhances the inflammatory cytokines secretion in BMDCs.
