摘  要:取出体质量0.12~0.15 kg的野生蒙古鲌Culter mongolicus肝脏和肠道,将肠道切分成前肠、中肠和后肠,在30℃下置于以0.2 mol/L的磷酸氢二钠-柠檬酸缓冲液调节pH为6.2、6.6、7.0、7.4和7.8下,采用考马斯亮蓝法和分光光度计法测定肝胰脏和肠道中蛋白酶、淀粉酶和脂肪酶的活性。结果表明,pH6.2~7.8时蒙古鲌肝胰脏、前肠、中肠、后肠中3种消化酶活性都呈先上升再下降的趋势;肝胰脏中各种消化酶活性在不同pH下差异性显著(P<0.05),肠道各部位中各种消化酶活性在不同pH下无差异性(P>0.05)。pH7.4时各部位的消化酶活性最高,蛋白酶活力(IU/g蛋白)强弱顺序为前肠(236.65)>肝胰脏(167.13)>中肠(62.64)>后肠(47.10);淀粉酶活力(IU/g蛋白)强弱顺序为肝胰脏(12.34)>前肠(3.17)>中肠(2.59)>后肠(2.28);脂肪酶活力(IU/g蛋白)强弱顺序为肝胰脏(186.74)>前肠(51.51)>中肠(37.69)>后肠(17.28)。结果表明肝胰脏是蒙古鲌消化酶的主要分泌场所,前肠为其主要消化部位。Activities of protease,amylase and lipase were investigated in hepatopancreas and intestine exposed to pH values of 6.2,6.6,7.0,7.4 and 7.8 in wild culter Culter mongolicus with body weight of 0.12~0.15 kg by methods of Coomassie brilliant blue and spectrophotometer at 30℃.Results showed that the activities of digestive enzymes in different parts of intestine at pH 6.2~7.8 were increased first and then decreased,with significant difference in hepatopancreas at different pH values(P<0.05),and without significant difference in different parts of the intestine at different pH values(P>0.05).There were the maximal activities of different digestive enzymes both in hepatopancreas and intestine at pH 7.4.The descending order of protease activity of digestive tract in different parts was expressed as foregut(236.65 IU/g),hepatopancreas(167.13 IU/g),midgut(62.64 IU/g)and hindgut(47.10 IU/g).However,the descending order of the amylase activity was described as hepatopancreas(12.34 IU/g),foregut(3.17 IU/g),midgut(2.59 IU/g)and hindgut(2.28 IU/g).There was higher lipase activity in hepatopancreas(186.74 IU/g)than that in foregut(51.51 IU/g),midgut(37.69 IU/g)and hindgut(17.28 IU/g).It is concluded from the above that the hepatopancreas is the most important site of digestive enzyme secretion,and the foregut is the main digestive site in culter Culter mongolicus.
