摘 要:目的:观察消渴平合剂对糖尿病肾病db/db小鼠肾脏NF-κB(核因子-κB)、IL-6(白介素-6)表达的影响,探讨其防治糖尿病肾病的机制。方法:选取24只db/db小鼠,随机分为模型组、厄贝沙坦治疗组、消渴平合剂治疗组,每组各8只,另取8只db/m小鼠作为正常对照组;各组小鼠分别连续给药8周后,检测各组小鼠糖化血清蛋白、三酰甘油、胆固醇、低密度脂蛋白、高密度脂蛋白的变化。留取肾组织,采用荧光定量PCR检测肾脏NF-κB p65mRNA、IL-6 mRNA的表达,Western blot检测肾脏NF-κB p65蛋白、IL-6蛋白的表达。结果:消渴平合剂可以明显降低db/db小鼠糖化血清蛋白水平,可明显降低db/db小鼠胆固醇、三酰甘油以及低密度脂蛋白水平,显著升高高密度脂蛋白水平。荧光定量PCR及Western blot结果显示,与模型组相比,消渴平治疗组显著降低NF-κB p65mRNA、IL-6 mRNA的表达(P<0.05),可明显抑制NF-κB p65蛋白、IL-6蛋白的表达(P<0.05)。结论:消渴平合剂可明显降低db/db小鼠糖化血清蛋白水水平,调节血脂,其机制可能与抑制肾脏NF-κB p65、IL-6基因和蛋白的表达有关。Objective:To observe the effects of Xiaokeping Mixture on the expressions of NF-κB and IL-6 in the kidney of db/db mice,and to explore the mechanism of its prevention and treatment of diabetic kidney disease.Methods:24 db/db mice were randomly divided into model group,Xiaokeping treatment group and Irbexatan group with 8 mice in each group,another 8 db/m mice were selected as the normal control group.Changes of glycosylated serum protein,triglyceride,cholesterol,low-density lipoprotein and high-density lipoprotein in mice of each group were detected after 8 weeks of continuous administration.Kidney tissues were collected and and the expressions of NF-κB p65 and IL-6mRNA in the kidney were detected by fluorescence quantitative PCR,the expressions of NF-κB p65 and IL-6 proteins in the kidney were detected by Western blot.Results:Xiaokeping Mixture could significantly reduce the level of glycosylated serum protein in db/db mice.It could significantly reduce cholesterol,triglyceride and low-density lipoprotein levels in db/db mice,and significantly increase high-density lipoprotein levels.Quantitative PCR and Western blot results indicated that,Xiaokeping Mixture could significantly reduce the expressions of NF-κB p65 and IL-6mRNA(P<0.05),inhibit the expressions of NF-κB p65 and IL-6 proteins(P<0.05),when compared with the model group.Conclusion:Xiaokeping Mixture can significantly reduce the glycosylated serum protein and regulate blood lipids in db/db mice,and the mechanism may be related to inhibiting the expression of NF-κB p65 and IL-6 genes and proteins in the kidney.
