森登-4味汤对膝骨性关节炎家兔关节软骨组织MMP-1、TIMP-1表达的影响

摘　　要：目的:观察蒙药森登-4味汤对膝骨性关节炎家兔膝关节关节面变化及软骨组织MMP-1(基质金属蛋白酶-1)、TIMP-1(基质金属蛋白酶抑制物-1)蛋白表达的影响,探讨蒙药森登-4味汤对膝骨性关节炎的治疗作用及作用机制。方法:实验分为正常组,模型组,森登-4味汤高、中、低剂量5组,每组16只。改良Hulth法进行动物造模,森登-4味汤给药4周后取材,进行膝关节关节面评分,采用Western blot检测关节软骨组织中MMP-1、TIMP-1蛋白表达的变化。结果:与正常组比较,模型组膝关节关节面评分增加,MMP-1、TIMP表达增高(P<0.05);与模型组比较,森登-4味汤各组关节面评分降低,MMP-1表达量降低,TIMP-1表达量升高(P<0.01或P<0.05)。结论:蒙药森登-4味汤对膝骨性关节炎模型家兔关节软骨组织MMP-1、TIMP-1蛋白表达的调节作用可能是其治疗膝骨性关节炎的作用途径之一。Objective:To observe the effects of Mongolian medicine Senden-4-Wei Decoction on the changes of knee joint surface and the expressions of MMP-1 and TIMP-1 protein in cartilage tissue of knee osteoarthritis rabbits,and to explore the therapeutic effect and mechanism of Mongolian medicine Senden-4-Wei Decoction on knee osteoarthritis.Methods:The experiment was divided into 5 groups:normal group,model group,high,medium and low dose of Senden-4-Wei Decoction groups,16 rats in each group.The modified Hulth method was used to establish animal models.After 4 weeks of Senden-4-Wei Decoction administration,the knee joint surface score was performed,and the expressions of MMP-1 and TIMP-1 protein in articular cartilage tissue were detected by Western blot.Results:Compared with the normal group,the knee joint surface score of the model group was increased,the expressions of MMP-1 and TIMP-1 were increased in the model group(P<0.05);Compared with the model group,the joint surface score of each Senden-4-Wei Decoction group was decreased,the expression of MMP-1 was decreased and the expression of TIMP-1 was increased(P<0.01 or P<0.05).Conclusion:The regulation effects of Mongolian medicine Senden-4-Wei Decoction on the expressions of MMP-1 and TIMP-1 protein in articular cartilage tissue of rabbits with knee osteoarthritis may be one of its therapeutic approaches.
