摘 要:环磷酸腺苷(cAMP)在微生物细胞内由ATP直接环化形成,嘌呤环的合成需要相关前体氨基酸的参与.为了探究添加不同前体氨基酸对cAMP发酵性能的影响,采用单因素试验和正交试验,确立前体氨基酸的最佳质量浓度配比.试验结果表明:当天冬氨酸、甘氨酸和谷氨酸的质量浓度分别为0.2 g/L、1.0 g/L和4.0 g/L时,cAMP产量达到最大为2.97 g/L.与对照1.42 g/L相比,cAMP产量提高了109.15%.依据正交试验结果,在7 L发酵罐上进行了验证.结果表明:当脉冲式添加氨基酸混合液发酵60 h时,cAMP的产量达3.35 g/L,相比于对照批次发酵72 h时cAMP质量浓度为3.32 g/L,发酵时间缩短了12 h,生产效率提高了21.7%.该结果说明添加氨基酸提高了cAMP的发酵性能.In live cells,cyclic adenosine monophosphate(cAMP)was produced from ATP catalyzed by adenylate cyclase and the synthesis of purine ring requires the participation of related precursor amino acids.In order to explore the effect of adding different precursor amino acids on cAMP fermentation performance,single factor experiment and orthogonal experiment were used to establish the optimal concentration ratio of precursor amino acids.The results showed that the maximal cAMP yield 2.97 g/L was achieved by coupling addition of aspartic acid,glycine,and glutamine with 0.2 g/L,1.0 g/L and 4.0 g/L respectively,which had increased by 109.15%compared to the control.Based on the orthogonal experiment results,the experiment was carried out on 7 L fermenter.The results showed that the yield of cAMP reached 3.35 g/L at 60 h by pulsed amino acid mixture fermentation.Compared with the control batch fermented for 72 h,the cAMP concentration was 3.32 g/L,the fermentation time was shortened by 12 h,and the production efficiency was increased by 21.7%.The results showed that the addition of amino acids improved the fermentation performance of cAMP.
