摘 要:目的探讨电针上巨虚和足三里穴对阳明腑实证肠运动功能的影响及可能的作用机制。方法60只Wistar大鼠随机分为空白组、模型组、电针组,每组20只。模型组与电针组连续灌服热性中药12天,第13、14天自身粪便混悬液灌胃,15天腹腔注射大肠杆菌菌液,建立阳明腑实证大鼠模型;空白组用等量生理盐水灌胃和腹腔注射。电针组大鼠于造模成功后每日电针双侧“足三里”和“上巨虚”穴,每次30 min,持续5天;空白组和模型组仅捆绑固定不干预。干预完成后检测大鼠小肠上段肌电慢波频率和振幅,评估小肠运动功能变化;取小肠上段组织标本进行HE染色观察小肠病理损伤;运用免疫荧光共聚焦显微镜、透射电镜观察肠神经胶质细胞形态及超微结构改变。结果与空白组比较,模型组大鼠小肠慢波频率减慢,振幅降低(P<0.05或P<0.01);小肠病理损伤明显,肠神经胶质细胞形态异常,网络结构紊乱,荧光强度OD值降低(P<0.05),透射电镜观察肠神经胶质细胞染色质聚集,分布不均,线粒体肿胀;电针组大鼠小肠慢波频率增快,振幅升高(P<0.05),小肠病理损伤及肠神经胶质细胞及网络结构损伤减轻,荧光强度OD值增加(P<0.05),肠神经胶质细胞超微结构损伤改善。结论电针“上巨虚”和“足三里”可促进阳明腑实证模型大鼠小肠运动功能恢复,其机制可能与修复肠神经胶质细胞网络损伤有关。Objective To investigate the effects and mechanism of electroacupuncture on Zusanli(ST36)and Shangjuxu(ST37)on intestinal motility of Yangming fu-viscera excess syndrome.Methods A total of 60 Wistar rats were randomly divided into a control group,a model group and an electro-acupuncture group,with 20 rats in each group.The model group and the electro-acupuncture group were continuously administrated with Chinese drugs of heat nature for 12 days.On the 13th and 14th day,the self-fecal suspension was intragastrically administered,and the E.coli bacterial liquid was injected intraperitoneally on the 15th day to establish a rat model of Yangming Fu-viscera excess syndrome;for the control group,the equal amount of normal saline was intragastrically and intraperitoneally injected.The rats in the electroacupuncture group were electro-acupunctured at bilateral“Zusanli(ST36)”and“Shangjuxu(ST37)”every day,30 minutes each time for 5 days after the successful modeling;the control group and the model group were only bound and fixed without intervention.After the intervention was completed,the frequency and amplitude of myoelectric slow wave in the upper small intestine of rats are detected to assess the changes in small intestine motor function;the upper small intestine tissue samples were taken for HE staining to observe the pathological damage of the small intestine;the enteric glial cells were observed by immunofluorescence confocal microscope and transmission electron microscope changes in morphology and ultrastructure.Results Compared with the control group,the small intestine slow wave frequency and amplitude of rats in the model group were slowed down(P<0.05 or P<0.01);small intestine pathological damage was obvious,enteric glial cell morphology was abnormal,network structure disorder,fluorescence intensity OD value decreased(P<0.05),transmission electron microscope observation showed enteric glial cell chromatin aggregation,uneven distribution,mitochondria swelling;in the electroacupuncture group,the small int
