摘 要:目的建立以苋菜红为荧光探针测定叶酸(folic acid,FA)含量的新方法。方法基于在pH=10.2的NH3-NH4Cl缓冲溶液中,FA能对苋菜红在420 nm处的荧光发射产生猝灭现象,且其猝灭程度与FA浓度呈良好线性关系,建立FA含量测定的新方法。考察了pH值、反应时间、FA浓度、共存物质和温度等因素对测定的影响,并分析其猝灭机制。结果 FA浓度在9.156×10^-6~9.156×10^-5 mol·L^-1范围时,其线性回归方程为F0/F=0.997 0+6 276 CFA,相关系数为0.999 4。在一定浓度范围内,葡萄糖、组氨酸、血清蛋白、可溶性淀粉及多种盐类等共存物质对检测体系的干扰都比较小。将此法用于2种规格FA片剂的含量测定,结果满意。经6次平行测定,该方法平均回收率分别为98.0%和96.6%,相对标准偏差分别为1.9%和1.3%。结论此法简单、快速、灵敏,为FA的含量测定提供了新的方法。OBJECTIVE To establish a new method for the determination of folic acid(FA) with amaranth as fluorescent probe. METHODS Based on the fact that FA could quench the fluorescence of amaranth at 420 nm in NH3-NH4Cl buffer solution(pH=10.2) and the degree of fluorescence quenching showed a good linear relationship with the concentration of FA,a new method for the determination of FA was established. The effects of pH value,reaction time,FA concentration,co-existing substances and temperature on the determination were investigated,and the quenching mechanism was analyzed. RESULTS When FA concentration ranged from 9.156×10^-6-9.156×10^-5 mol·L^-1,the linear regression equation was F0/F=0.997 0+6 276 CFA,and the correlation coefficient was 0.999 4. Within a certain concentration range,glucose,histidine,serum protein,soluble starch and various salts and other co-existing substances had little interference to the determination of FA. The method was applied to the content determination of FA tablets with two specifications and the results were satisfactory. The average recoveries were 98.0% and 96.6%,and the RSD was 1.9% and 1.3%,respectively. CONCLUSION The method is simple,rapid and sensitive,which provides a new method for the determination of FA.
